黄瓜花叶病毒在甜瓜种子中的带毒检测及脱毒方法研究 被引量:3

摘要 为明确甜瓜种子携带黄瓜花叶病毒(Cucumher mosaic virus,CMV)状况,以5个品种甜瓜种子为材料,采用实时荧光定量PCR技术检测甜瓜种子、胚芽中CMV带毒率和带毒量。结果表明,供试种子、种胚带毒率分别为80%~96.6%和5.0%~25.0%,带毒量分别为6.62×10^4~5.47×10^7拷贝/粒和3.78×10^4~1.00×10^6拷贝/粒;种苗传毒率为0~6.6%,不同品种之间存在差异。种子的带毒率高而传毒率低,表明CMV主要存在于甜瓜种子的种皮上。试验比较了5种处理方法对带CMV甜瓜种子的脱毒效果。干热(65℃)、湿热(100℃沸水)、温汤(55℃)、干热+湿热(65℃干热+100℃沸水)、干热+10%Na 3PO4(65℃干热+10%Na3PO4)处理甜瓜种子,均能降低种子带毒量。干热、湿热、干热+湿热和干热+10%Na3PO4处理后种子脱毒率可达96%以上,显著优于温汤处理(P<0.05),并能促进种子发芽。干热+10%Na 3PO4处理甜瓜种子后的种苗中未检出病毒,对CMV的脱除效果最佳。 The study aims to determine melonseeds infected by Cucumber mosaic virus(CMV),the infection rate of seed,plumule,and seed-to-seedling transmission of CMV in five melon varieties were detected,and the effect of five treatments on the virus elimination were evaluated by using fluorescence real-time RT-PCR method.The results showed that the infection ratesof seeds and plumule infected by CMVwere 80%-96.6%and 5.0%-25.0%,the amounts of the virus were 6.62×10^4-5.47×10^7 and 3.78×10 4-1.00×10^6 copies/one seed,respectively.The seed-to-seedling transmission ratesbetween 0-6.6%were different depended on the varieties of melon seed.The transmission rate of seed-to-seedling was low while the infection rate was high,this indicated that the virus mainly distributed on the surface of the seeds.The effects of five treatments(65℃dry heat,100℃wet heat,55℃wet heat,65℃dry heat+100℃wet heat,65℃dry heat+10%Na 3PO 4)on virus elimination varied.The elimination efficiency of CMV was more than 96%when the seeds were treated by 65℃dry heat,100℃wet heat,65℃dry heat+100℃wet heat and 65℃dry heat+10%Na 3PO 4,also these treatments could promote the seed germination.The treatment of dry heat and 10%Na 3PO 4 showed that the virus detection rate of the seedling was zero,which was the most effective method for removing CMV from the melon seeds.
