滴鼻感染铜绿假单胞菌致大鼠肺炎模型的评价 被引量:6

摘要 目的:客观评价已建立的滴鼻途径感染致肺炎大鼠模型,为后续肺炎防治机制研究提供实验依据。方法:将24只SD雄性大鼠按随机数表法分为对照组和实验组,每组12只,两组均按干预后0d、5d、10d和15d分为4个亚组,每组3只。以滴鼻途径干预,采集干预0d、5d、10d和15d的各亚组肺泡灌洗液(BALF)及肺组织样本,检测其肺指数、BALF中白细胞数、免疫炎性因子IL-4、TNF-α和IFN-γ的表达及铜绿假单胞菌数量,并分别与对照组的相应亚组比较分析。结果:实验组大鼠肺指数在造模干预后,随时间延长5d、10d和15d逐渐增大,与对照组相比差异有统计学意义(t=-2.46,t=-3.26,t=-3.73;P<0.05);BALF中白细胞数均明显高于相应对照亚组,BALF中IL-4和TNF-α在造模干预后均呈增长趋势,两组比较差异有统计学意义(t=4.24,t=4.88;P<0.05),IFN-γ则呈下降趋势;BALF中铜绿假单胞菌数在造模干预后,随时间延长呈增多趋势。结论:成功构建的滴鼻感染耐药铜绿假单胞菌致大鼠肺炎模型,可用于中医药防治肺炎的机制研究。 Objective: To evaluate objectively the established rat pneumonia model caused by intranasal inoculation so as to provide experimental basis for the followed mechanism study of prevention and treatment of pneumonia. Methods: 24 SD male rats were divided into control group (n=12) and experimental group randomly (n=12), and each group was further divided into 4 subgroups as 0, 5, 10 and 15 days post intervention, with 3 rats in each subgroup. The rats in all groups were intervened by intranasal inoculation, and their lung tissue samples and bronchoalveolar lavage fluid (BALF) were collected from every sub-group at 0, 5, 10 and 15 days post intervention. And then, the lung index, the white blood cell count, IL-4, TNF-α, IFN-γ and the amount of pseudomonas aeruginosa (PA) in BALF were detected. And the results of different subgroups of two group were further compared. Results: After intervention, the lung index of the experimental group increased gradually with the time prolonged (from 5d to 15d), which was different to that of control group significantly (t=-2.46, t=-3.26, t=-3.73, P<0.05). The number of white blood cell in BALF of the experimental sub-group were significantly higher than those of control sub-group. And IL-4 and TNF-αin in BALF of experimental group appeared growing trend after molding intervention, and the differences of them between the two groups were significant (t=4.24, t=4.88, P<0.05), while IFN-γ was opposite. The number of PA in BALF of the experimental group appeared increasing trend with time extension after molding intervention. Conclusion: The rat pneumonia model are constructed successfully from PA by intranasal inoculation can be used in the mechanism study of prevention and treatment with traditional Chinese medicine (TCM) for pneumonia.
