摘要
本文建立了高效液相色谱法测定婴幼儿配方奶粉中维生素A和维生素E的含量。方法:称取2~5 g奶粉样品,温水溶解后加入乙醇和氢氧化钠溶液后皂化,正己烷萃取,正己烷定容,进样量为25μL,以正己烷∶四氢呋喃(90∶10)为流动相,流速为1.5 mL/min,在Agilent Zorbax RX-SIL,3μm,4.6×150 mm硅胶柱中分离,在紫外波长325 nm下测定维生素A,294 nm下测定维生素E。结果:维生素E的检出限为0.07~0.14 mg/kg,维生素E的定量限为0.12~0.4 mg/kg,维生素A的检出限为0.02 mg/kg,维生素A的定量限为0.04 mg/kg,在范围内线性良好,相关系数大于0.99,回收率在80%~100%之间。结论:该方法分离度佳,定量准确,能满足婴幼儿配方奶粉中维生素A和四种维生素E异构体的检测需求,适用于婴幼儿配方奶粉维生素A和维生素E的放行检测。
A high-performance liquid chromatography(HPLC) method has been established for the determination of vitamin A and four isomers of vitamin E in infant formula milk powder.The samples are analyzed by HPLC use Agilent Zorbax RX-SIL silica gel column(4.6×150 mm,3 μm). The mobile phase is a mixture of hexane and tetrahydrofuran(90∶10,v∶v) at a flow rate of 1.5 mL/min. The column temperature is 40℃,vitamin A is determined at 325 nm,vitamin E is determined at 294 nm. Results: LOD of vitamin E is 0.07~0.14 mg/kg,LOQ of vitamin E is 0.12~0.4 mg/kg,LOD of vitamin A is 0.02 mg/kg,LOQ of vitamin A is 0.04 mg/kg. The linearity is good in the range,the linear correlation coefficient is more than 0.99,and the recovery rate is between 80%~100%. Conclusion: The method has good separation and accurate quantification,it can meet the needs of the determination of vitamin A and four isomers of vitamin E in infant formula milk powder. It is suitable for the release detection of vitamin A and vitamin E in infant formula milk powder.
