摘要
目的:比较高卢蜜环菌M1、M2两菌株胞外酶活性、菌丝多糖变化规律及菌株生长量,初步筛选出优良菌株。方法:利用改良PDA固体培养基培养菌株M1和M2,采用十字交叉法测定生长量,ABTS法及DNS显色法检测胞外酶活性,用浓硫酸-苯酚法对二者多糖含量进行测定。结果:M1菌株生长速度快,菌索粗壮,分支较多,长势较好。木聚糖酶是蜜环菌主要作用的胞外酶,其活力远高于纤维素酶、漆酶和果胶酶,M1菌株的木聚糖酶活力更高。M1和M2菌丝体多糖含量最高分别为38.958 mg/g和32.717 mg/g,多糖的累积与其分泌的胞外酶漆酶、木聚糖酶活力呈现显著相关性。结论:通过对M1、M2两菌株生长量、胞外酶活力及菌丝体多糖的比较和分析,初步认为M1为优势菌株。
Objective:To select a better stain from two Armillaria gallica strains by comparing the growth of the strains,extracellular enzymatic activities and changes of the polysaccharides content.Methods:The two strains,M1 and M2,were cultured with modified PDA solid culture medium.The growth of the strains was measured by cross method.And the chromogenic method of ABTS and DNS were used to measure the extracellular enzyme activities.The content of polysaccharide was determined by concentrated sulfuric acid-phenol method.Results:The growth rate of M1 strain was faster than that of M2 strain.The rhizomorph of M1 strain was thicker and stronger than that of M2 strain,with more branches and better growth.Xylanase was the major extracellular enzyme of Armillaria gallica and its activity was much higher than that of CMC enzyme,laccase and pectinase.The xylanase activity of M1 strain was higher than that of M2.The highest polysaccharide content of M1 and M2 mycelia was 38.958 mg/g and 32.717 mg/g,respectively.The accumulation of polysaccharide of Armillaria gallica rhizomorph showed a significant correlation with the activities of extracellular enzymes laccase and xylanase.Conclusion:After the comparison and analysis of the two stains,M1 is the dominant strain.
