摘要
目的:研究四逆散对慢性萎缩性胃炎癌前病变大鼠的作用,为更好地应用于临床提供依据。方法:采用MNNG造模法复制胃黏膜上皮异型增生模型,以不同浓度四逆散处理大鼠,观察病变大鼠体征、体重、脏器系数、胃黏膜组织的变化,采用ELISA法测定PGⅠ、PGⅡ、G-17含量,Western Blot法检测PCNA、Bcl-2、Bax蛋白表达。结果:四逆散高剂量组体重增长最缓,低、中剂量组对大鼠脏器无明显影响,但高剂量组与正常组相比在肾脏系数,与模型组相比在胸腺系数上有一定差异;四逆散中、高剂量组能降低PGⅡ、升高PGR,四逆散高剂量组能升高G-17浓度;四逆散能降低病变大鼠胃黏膜组织PCNA、Bcl-2的表达,升高Bax的蛋白量。结论:四逆散能抑制或逆转慢性萎缩性胃炎癌前病变的发生,可能与调控细胞增殖与凋亡平衡有关。
Objective:To study the effect of Sinisan on the rats of precancerous lesion of chronic atrophic gastritis,and provide scientific basis for better clinical application. Methods:The model rats of gastric mucosa epithelial hyperplasia were replicated by MNNG,then different concentrations of Sinisan treated them,observed physical signs,body weight,viscera coefficient,gastric mucosal tissue,detected the PG Ⅰ,PG Ⅱ,G-17 content by ELISA,expressions of PCNA,Bcl-2,Bax were examined by Western blot. Results:The group of high dose Sinisan got the slowest pace in weight,low and medium dose of Sinisan had no obvious effect on rat organs,but kidney coefficient of high dose group was different from the control group,thymus coefficient of high dose group was different from the model group. Medium and high dose of Sinisan could decrease PGⅡand increase PGR,high dose group of Sinisan could increase the concentration of G-17;compared with model group,PCNA and Bcl-2 protein decreased with Sinisan treated,Bax took on a uptrend . Conclusion:To some extent,Sinisan can inhibit or reverse the occurrence of precancerous lesions of chronic atrophic gastritis,maybe the regulatory mechanism is related to balance proliferation and apoptosis.
