33份节瓜育种材料的遗传多样性ISSR分析 被引量:5

Genetic Diversity of 33 Chieh-qua Resources Based on ISSR

摘要 通过比较前人在节瓜遗传多样性研究的方法,本研究选择ISSR分子标记技术对33份节瓜育种材料的遗传多样性进行分析。首先利用Tm值的差异将100条ISSR引物进行分组,经筛选得到适合本次研究的15条ISSR引物。然后通过ISSR-PCR扩增,2%琼脂糖凝胶电泳,UVP成像,共得到61条条带,其中多态性的条带占55条,多态性的比例为90.16%。通过聚类分析得到33份节瓜材料的遗传相似系数介于0.52~0.95之间,说明多态性位点较多,这些节瓜材料的遗传差异较大。当遗传相似系数为0.73时,这些节瓜材料分成5个居群,节瓜材料J18-11与其它材料的相似性系数为0.52,说明节瓜材料J18-11与其它材料遗传差异较大。通过对这33份节瓜材料的遗传多样性分析,明确了这些材料之间的遗传差异与相似性程度,对后期的育种工作亲本选择以及基因连锁图谱的构建提供了参考。 ISSR markers were used for the study of genetic diversity of 33 chieh-qua inbred lines because ISSR is the more suitable for the analysis of genetic diversity of chieh-qua than other methods adopted previously. Firstly,all of the primers were divided into two groups based on Tm values. Then 15 obviously polymorphic primers were selected for the next test. After the PCR amplification, agarose gel electrophoresis and UVP imaging, totally 61 fragments and 55 polymorphic fragments were obtained and the polymorphism ratio was 90.16%. The molecular genetic similarity coefficient ranged from 0.52 to 0.95 through cluster analysis, which indicated that there were many polymorphic sites and much genetic difference of these chieh-qua materials. When the similarity coefficient was 0.73, these chieh-qua materials were divided into 5 clusters. The smallest similarity coefficient was 0.52, which showed the difference between J18-11 and other members in genetics. In conclude, it was clarified that the difference and similarity of 33 chieh-qua inbred lines in genetics, which was provided guides for chieh-qua breeding and parents selection in constructing genetic linkage map of chieh-qua.
