摘要
目的:探讨“白芍-桂枝”药对治疗类风湿关节炎的作用机制。方法:从中药系统药理学数据库和分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)中寻找与“白芍-桂枝”药对有关的所有化学成分,选择口服生物利用度≥30%、类药性≥0.18作为化合物分子的筛选条件;通过TCMSP寻找与候选化合物相关的潜在靶点,构建药物靶点相互作用的网络图和靶点疾病相互作用的网络图。通过治疗靶点数据库查找类风湿关节炎相关靶点,综合分析得到化合物与疾病相关靶点,构建网络图。基于京都基因与基因组百科全书的生物通路富集分析,研究“白芍-桂枝”药对治疗类风湿关节炎的作用机制。结果:与类风湿关节炎相关的“白芍-桂枝”药对“活性成分-潜在靶点”网络包含32个节点;涉及的关键靶点有白细胞介素6、肿瘤坏死因子等。结论:本研究结果初步验证和预测了“白芍-桂枝”药对治疗类风湿关节炎的作用机制,为进一步深入揭示其作用机制奠定了良好的基础。
OBJECTIVE:To probe into the mechanism of “Paeoniae Radix Alba-Cinnamomi Ramulus” in the treatment of rheumatoid arthritis.METHODS:All the chemical constituents related to “Paeoniae Radix Alba-Cinnamomi Ramulus” were searched from traditional Chinese medicine systems pharmacology database and analysis platform (TCMSP).Oral bioavailability (OB)≥30% and pharmacokinetic (DL)≥0.18 were selected as the screening conditions for compounds.The potential targets related to candidate compounds were identified by TCMSP,and then the network diagrams of drug-target interaction and disease-target interaction were constructed.Therapeutic Target database (TTD) database was used to find rheumatoid arthritis related targets,comprehensive analysis was conducted to obtain compounds and disease related targets,and the network was established.Based on the enrichment analysis of biological pathway in Kyoto Encyclopedia of Gene and Genome (KEGG),the mechanism of “Paeoniae Radix Alba-Cinnamomi Ramulus” in the treatment of rheumatoid arthritis was studied.RESULTS:The active component-potential target network of “Paeoniae Radix Alba-Cinnamomi Ramulus” related to rheumatoid arthritis contained 32 nodes.The key targets involved were interleukin-6 (IL-6) and tumor necrosis factor (TNF).CONCLUSIONS:The results of this study preliminarily validate and predict the mechanism of “Paeoniae Radix Alba-Cinnamomi Ramulus” in the treatment of rheumatoid arthritis,and lay a solid foundation for further revealing its mechanism.
