摘要
该研究以米曲霉(Aspergillus oryzae)ZJGS-LZ-12为研究对象,米曲霉3.042为对照,通过双向电泳结合生物质谱检测,对其胞外分泌蛋白差异进行比较分析。结果表明,与米曲霉3.042相比,米曲霉ZJGS-LZ-12胞外蛋白的分泌能力相当,但胞外分泌蛋白组存在显著差异(P<0.05)。差异表达蛋白点共169个,其中80个蛋白点分泌表达量下调,89个蛋白点分泌表达量上调。差异蛋白质主要参与糖类和蛋白质水解过程,主要是水解淀粉、纤维素、半纤维素及蛋白质的酶。其中,淀粉水解酶、半纤维素酶活性显著增强,β-1,4-内切木聚糖酶F3、β-半乳糖苷酶A和阿拉伯半乳聚糖-β-内切半乳聚糖酶A分泌表达量分别上调292、1 156和2 569倍,蛋白酶Pep1和Alp1分泌表达量分别上调100和1 300倍,充分表明运用此菌株发酵有利于改善豆酱的风味和品质。
Using Aspergillus oryzae ZJGS-LZ-12 as the research object, A. oryzae 3.042 as the control, the difference of extracellular secreted proteins was compared and analyzed by two-dimensional electrophoresis combined with mass spectrometry. The results showed that compared with A. oryzae 3.042, the secretion ability of extracellular protein for A. oryzae ZJGS-LZ-12 was comparable, but there was significant difference in extracellular secretome (P<0.05). The differential protein spots were 169, among them, the expression quantities of the 80 protein spots were down-regulated, and the 89 protein spots were up-regulated. These differential proteins were mainly involved in the hydrolytic process of saccharides and proteins, and they were mainly the enzyme that hydrolyze starch, cellulose, hemi-cellulose and protein. Among them, the amylolytic enzyme and hemicellulase activities were significantly enhanced, the expression quantities of the β-1,4-endoxylanase F3,β-galactosidase A and arabinogalactan-endo-β-galactanase A were up-regulated 292, 1 156 and 2 569 times in the A. oryzae ZJGS-LZ-12, respectively. And the expression quantities of protease Pep1 and Alp1 were up-regulated 100 and 1 300 times respectively, indicating that the fermentation of the strain was beneficial to improve the flavor and quality of soybean paste.
