牛传染性鼻气管炎病毒的分离鉴定及其遗传进化分析 被引量:7

Isolation,identification and genetic evolution analysis of an infectious bovine rhinotracheitis virus strain

摘要 采集内蒙古地区疑似发生牛传染性鼻气管炎的发病牛的鼻拭子液,经MDBK细胞连续传代培养,分离获得1株病毒,该病毒在MDBK细胞上增殖致细胞病变。经PCR鉴定、间接免疫荧光试验和动物回归试验,证明该分离株为牛传染性鼻气管炎病毒(IBRV),对牛具有较强的致病性,将其命名为IBRV NM8株。通过PCR扩增分别获得gB和gE全基因序列,遗传进化分析表明,IBRV NM8株g B和g E基因与BHV-1型参考毒株的同源性较高,位于同一进化分支,而与BHV-5型参考毒株位于不同的进化分支。NM8株gB基因与Cooper株、MN12株、Los Angeles株和NM06株等的亲缘关系相对较近,而g E基因与瑞典的Salwa株的亲缘关系较近,位于同一分支。上述研究结果将为我国IBRV流行病学研究提供新的数据,并为相关后续研究奠定基础。 Nasal swabs were collected from cattle with suspected infectious bovine rhinotracheitisin Inner Mongolia and passaged continuously in MDBK cells.Then one virus strain that can cause cytopathic effects was obtained.The strain was confirmed to be infectious bovine rhinotracheitis virus(IBRV) by PCR,indirect immunofluorescence assay and animal infection test,which has strong pathogenicity to cattle,and it was named as IBRV NM8 strain.The gB and gE genes were amplified by PCR,and the phylogenetic analysis showed that the IBRV NM8 strain has a high homology with the BHV-1 reference strains and belong to the same branch,and it was in the different branch with BHV-5.The gB gene of NM8 strain is relatively close to Cooper,MN12,Los Angeles and NM06 strain,whilethe gE gene is relatively close to Swedish Salwa strain.These results provided new data for IBRV epidemiological research in China and lay a theoretical foundation for relevant research.
