摘要
目的探讨长链非编码RNA(lncRNA)ATB联合基质金属蛋白酶-1(MMP-1)与血清淀粉样蛋白A(SAA)对食管癌的诊断价值,及三者与食管癌临床病理特征的关系。方法收集食管癌患者135例,与同期行健康体检的健康人群150例。采用实时荧光定量PCR(RT-PCR)法检测受试者外周血中lncRNA ATB;采用酶联免疫吸附法(ELISA)和化学发光法分别检测MMP-1与SAA水平。结果食管癌患者lncRNA ATB、MMP-1与SAA水平显著高于健康人群,且差异具有统计学意义(P<0.05)。LncRNA ATB联合MMP-1与SAA在区分食管癌与健康人群的曲线下面积(AUC)为0.887(95%CI:0.822~0.951,P<0.001),灵敏度为79.7%,特异度为92.8%。此外,食管癌患者lncRNA ATB、MMP-1与SAA水平与肿瘤TNM分期、组织学类型、肿瘤分化程度等显著相关(P<0.05),而与患者性别、年龄、肿瘤部位等因素无关(P>0.05)。多因素分析显示,高lncRNA ATB、MMP-1与SAA水平是食管癌发病的独立危险因素,对食管癌发病风险的预测具有指导意义。结论lncRNA ATB、MMP-1与SAA对食管癌具有诊断价值,且与食管癌的TNM分期、组织学类型、分化程度显著相关。此外,lncRNA ATB、MMP-1与SAA亦可作为食管癌发病风险预测的参考指标。
Objective To explore the diagnostic values of long non-coding RNA(lncRNA)ATB,matrix metalloproteinase-1(MMP-1)and serum amyloid protein A(SAA)for esophageal cancer,and the relationship between the three and the clinicopathological features of esophageal cancer.Methods 135 cases of esophageal cancer were collected,and 150 healthy people were recruited during the same period.The levels of lncRNA ATB in the peripheral blood of subjects were detected by real-time quantitative PCR(RT-PCR).Enzyme-linked immunosorbent assay(ELISA)and chemiluminescence were used to detect MMP-1 and SAA levels,respectively.Results Plasma lncRNA ATB,MMP-1 and SAA levels in patients with esophageal cancer were significantly higher than those of healthy subjects,and the difference were statistically significant.When combined with lncRNA ATB,MMP-1 and SAA,the area under the curve(AUC)to distinguish between esophageal cancer and healthy subjects was 0.887(95%CI:0.822~0.951,P<0.001),the sensitivity was 79.7%,specificity was 92.8%.In addition,the expressions of lncRNA ATB,MMP-1 and SAA in plasma of patients with esophageal cancer were significantly correlated with TNM stage,histology classification and differentiation degree of tumor,but not related to gender,age,and tumor sites.Multivariate analysis showed that plasma lncRNA ATB,MMP-1 and SAA levels were both independent risk factors for esophageal cancer,which is of guiding significances for predicting the risk of esophageal cancer.Conclusion LncRNA ATB,MMP-1 and SAA have diagnostic values for esophageal cancer,and are significantly correlated with the differentiation degree,histology classification,and TNM stage of esophageal carcinoma.In addition,LncRNA ATB,MMP-1 and SAA can also serve as reference index for predicting the risk of esophageal cancer.
