摘要
目的观察糖痛方对糖尿病周围神经病变(DPN)大鼠坐骨神经自噬相关蛋白Beclin-1和LC3Ⅱ表达的影响,初步探讨其作用机制。方法雄性SD大鼠75只,随机选取15只为正常组,其余60只采用2%链脲佐菌素诱导制作糖尿病大鼠模型,造模成功4周后,结合缺血再灌注制备DPN模型,成模大鼠随机分为模型组和糖痛方低、中、高剂量组,每组15只,糖痛方低、中、高剂量组分别按成人6.25、12.5、25倍剂量给药,正常组和模型组给予等量蒸馏水灌胃,每日1次,灌胃8周,Western blot和实时荧光定量PCR分别检测大鼠Beclin-1、LC3Ⅱ蛋白和mRNA的表达,透射电镜观察坐骨神经超微结构。结果与正常组比较,模型组大鼠Beclin-1、LC3Ⅱ蛋白和mRNA表达均升高,髓鞘肿胀、变性,板层分离;与模型组比较,糖痛方各剂量组大鼠Beclin-1和LC3Ⅱ蛋白和mRNA表达呈下降趋势,以糖痛方中、高剂量组降低明显,髓鞘变性程度减轻。结论糖痛方可能通过抑制模型大鼠过度自噬,发挥对DPN大鼠的治疗作用。
Objective To observe the effects of Tangtong Prescription on beclin-1 and LC3Ⅱof autophagy related proteins in sciatic nerve of diabetic peripheral neuropathy rats;To preliminarily investigate its mechanism of action.Methods Seventy-five male SD rats were randomly selected,15 for normal group,and other 60 rats were treated with intraperitoneal injection of STZ to build type 2 diabetic nice model.4 weeks after modeling,diabetic peripheral neuropathy(DPN)model was prepared by combining with ischemia-reperfusion preparation of,the model of DPN rats were randomly divided into model group,and Tangtong Prescription low-,medium-and high-dosage groups,with 15 cases in each group.Tangtong Prescription low-,medium-and high-dosage groups were given medicine as 6.25,12.5,25 times of adult doses.The normal group and the model group were given the same amount of distilled water for gavage,once a day.After gavage for 8 weeks,Western blot and real-time fluorescent quantitative PCR were used to detect the expressions of Beclin-1 and LC3Ⅱproteins and mRNA,and the ultrastructure of sciatic nerve was observed by transmission electron microscope.Results Compared with the normal group,expressions of Beclin-1 and LC3Ⅱprotein and mRNA in the model group increased,with myelin sheath swelling and degeneration,and lamellar separation.Compared with the model group,expressions of beclin-1 and LC3Ⅱprotein and mRNA showed a downward trend,and myelin degeneration was significantly reduced in Tangtong Prescription medium-and high-dosage groups.Conclusion Tangtong Prescription may play a therapeutic role in DPN rats by inhibiting excessive autophagy in model rats.
