摘要
目的:通过比较TRIzol法、RNAeasy动物RNA抽提试剂盒法和HP Total RNA Kit试剂盒法3种常规的RNA提取方法,筛选获得其中提取药用昆虫九香虫总RNA最优方法。方法:分别采用3种常规的方法提取九香虫总RNA,并用1%琼脂糖凝胶电泳和NanoDrop 2000/2000C分光光度计检测总RNA的完整性、纯度及含量。结果:TRIzol法和HP Total RNA Kit试剂盒法提取RNA的完整性较好;紫外分光光度计结果显示3种方法提取九香虫总RNA的OD 260/280的值都在1.9~2.1,纯度较高;TRIzol法提取的RNA的含量远高于另两种方法提取的RNA的含量。结论:3种方法中,TRIzol法提取九香虫RNA的效果最好,是提取九香虫RNA较为可取的方法。
Objective:The TRIzol method,RNAeasy animal RNA extraction Kit method and HP Total RNA Kit method were compared for RNA extraction,to screen and obtain an optimal method for extracting total RNA from Aspongopus chinensis Dallas.Methods:The total RNA of Aspongopus chinensis Dallas was extracted by three methods.The integrity,purity and concentration of total RNA were detected by 1% agarose gel electrophoresis and NanoDrop 2000/2000 C spectrophotometer.Results:TRIzol method and HP Total RNA Kit had better efficiency in extracting RNA;UV spectrophotometer results showed that the OD260/280 values of total RNA extracted by three methods was range from 1.9 to 2.1,with high purity;the concentration of RNA extracted by TRIzol method was much higher than that of RNA extracted by the other two methods.Conclusion:TRIzol method is an effective method to extract the RNA of Aspongopus chinensis Dallas,and it is one of the preferable methods to extract the RNA of Aspongopus chinensis Dallas.
