丹皮酚对LPS诱导小鼠子宫内膜炎NF-κB信号通路的影响 被引量:15

Paeonol attenuates LPS-induced endometritis in mice via inhibiting NF-κB signal pathway

摘要 目的研究丹皮酚对LPS诱导小鼠子宫内膜炎的作用及其作用机制。方法60只小鼠随机分为假手术组、模型组、醋酸地塞米松组(5 mg/kg)、丹皮酚高剂量组(50 mg/kg)、丹皮酚中剂量组(25 mg/kg)、丹皮酚低剂量组(12.5 mg/kg),各组连续灌胃给药5 d。末次给药1 h后4%水合氯醛腹腔注射麻醉(10 ml/kg)后,打开腹腔,露出左侧子宫,除假手术组外,其余组用1 ml胰岛素注射器向子宫内注射(2.5μg/μl)LPS生理盐水25μl,假手术组注射25μl生理盐水。注射24 h后脱颈椎处死小鼠,分离子宫待测。HE染色法检测子宫组织病理变化,Griess法测定NO含量,ELISA法检测炎性因子MPO、IL-1β、TNF-α含量,Western blot法检测子宫组织P65/p-P65及IκBα/p-IκBα蛋白表达变化。结果研究结果显示丹皮酚能明显减轻LPS诱导的小鼠子宫内膜病理损伤,显著降低小鼠子宫组织MPO、NO、IL-1β、TNF-α含量,进一步研究结果显示丹皮酚可抑制小鼠子宫组织NF-κB p65、IκB蛋白的磷酸化水平。结论丹皮酚可通过抑制NF-κB信号通路、减少炎症细胞因子的释放从而发挥抗脂多糖诱导的小鼠子宫内膜炎的作用。 To investigate the effects of paeonol on LPS-induced mouse endometritis and its potential mechanism,60 mice were recruited and randomly divided into sham operation group,model group,dexamethasone acetate group(5 mg/kg),paeonol high dose group(50 mg/kg),paeonol medium dose group(25 mg/kg)and paeonol low dose group(12.5 mg/kg).Corresponding drug were administrated orally to mice for 5 days.One hour after the last administration,4%chloral hydrate was injected intraperitoneally(10 ml/kg),then the abdominal cavity was opened and the left uterus was exposed.Except for the sham-operated group,the other groups were injected with 25μl LPS saline(2.5μg/ml)into uterus using 1 ml insulin syringe,while the sham-operated group was injected with 25μl saline.Twenty four hours after the LPS injection,mice were sacrificed and uterus was separated for testing.Histopathological changes of uterus were detected by HE staining,NO content by Griess method,MPO,IL-1βand TNF-αcontent by ELISA method.The changes of P65/p-P65 and IκBα/p-IκBαproteins were detected by Western blotting.Data showed that paeonol could significantly alleviate LPS-induced endometrial pathological damage in mice,and significantly reduce the contents of MPO,NO,IL-1βand TNF-αin mouse uterus.Furthermore,paeonol could significantly suppressed LPS-induced phosphorylation of NF-κB p65 and IκBα.Taken together,Paeonol has protective effects against LPS-induced endometritis,which may be attributed to the suppression of inflammatory response,and inhibition of NF-κB signaling pathway.
