Determination of Alkaloids and Industial Dyestuffs in Cooked Meat by High Performance Liquid Chromatography Tandem Mass Spectrometry

摘要 为建立高效液相色谱-串联质谱法(High performance liquid chromatography-tandem mass spectrometry,HPLC-MS/MS)同时测定熟肉中5种罂粟壳生物碱和8种工业染料的方法,样品用60%乙腈提取,经氯化钠盐析后,有机相提取液经分散固相吸附剂净化,用乙腈和0.01 mol/L甲酸铵(含0.1%甲酸)溶液作为流动相进行梯度洗脱,MG II色谱柱进行色谱分离,经高效液相色谱-质谱/质谱电喷雾电离多反应监测正离子模式测定。13种化合物在各自的线性范围内呈现良好的线性,相关系数均大于0.99,检出限为0.030~0.75μg/kg,定量限为0.10~2.5μg/kg,加标浓度范围在0.10~25μg/kg之间,平均回收率为64.9%~108.8%,RSD为2.5%~10.7%(n=6)。该方法操作简单、准确快速,可应用于熟肉制品中罂粟壳生物碱和工业染料的测定。 A method is established for simultaneous determination of 5 kinds of poppy shell alkaloids and 8 kinds of industrial dyes in cooked meat products by high performance liquid chromatography tandem mass spectrometry(HPLC-MS/MS).The sample is extracted with 60%acetonitrile and salted out by sodium chloride,the organic phase extraction solution is purified by dispersive solid phase adsorbent,and gradient elution was performed with acetonitrile and 0.01 mol/L ammonium formate(containing 0.1%formic acid)solutions as mobile phase.The sample is separated by MG II chrmoatographic column,and determined by high performance liquid chromatography tandem mass spectrometry with multiple reaction monitoring and positive ion mode.The calibration curve of all 13 compounds shows a good linearity in the respective linear range,the correlation coefficients were all greater than 0.99,detection limit is 0.030-0.75μg/kg,the method limits of quantify are between 0.10μg/kg and 2.5μg/kg,the range of standardized concentration is 0.10-25μg/kg,the average standard addition recoveries are between 64.9%and 108.8%,and the relative standard deviation(RSD)are between 2.5%and 10.7%(n=6).The method is simple,accurate and rapid,and it can be used for the determination of poppy shell alkaloids and industrial dyes in cooked meat products.
