藏红花素对心肌微血管内皮细胞缺氧复氧损伤的保护作用研究 被引量:8

Protective Effect of Crocin Against Hypoxia/Reoxygenation Injury of Cardiac Microvascular Endothelial Cells

摘要 目的探讨藏红花素通过降低氧化应激水平对心肌微血管内皮细胞缺氧复氧损伤的保护作用。方法分离8周龄雄性C57BL/6N小鼠左心室心肌微血管内皮细胞(CMECs)。细胞分为4组:正常细胞组(Control组)、溶剂对照组(Vehicle组)、缺氧复氧组(H/R组)、缺氧复氧+藏红花素组(H/R+Crocin组)。检测细胞内超氧阴离子水平、细胞增殖能力、细胞迁移能力及Nox2水平。结果(1)与正常细胞组比较,缺氧复氧组CMECs细胞增殖能力降低(P<0.05),细胞迁移能力显著减弱、细胞内超氧阴离子水平显著升高(P<0.01);(2)予藏红花素预处理后,较缺氧复氧组,缺氧复氧+藏红花素组CMECs细胞增殖能力升高、细胞迁移能力增强、细胞内超氧阴离子水平降低(P<0.05);(3)Western Blot法显示,较正常细胞组,缺氧复氧组Nox2水平显著升高(P<0.01),予藏红花素预处理后,CMECs细胞经过缺氧复氧后,细胞内Nox2水平较缺氧复氧组降低(P<0.05)。结论藏红花素通过降低CMECs细胞缺氧复氧后Nox2水平,减少CMECs细胞内ROS水平,起到保护CMECs细胞缺氧复氧损伤的作用。 OBJECTIVE To investigate the protective effect of crocin against Hypoxia/Reoxygenation(H/R)injury of cardiac microvascular endothelial cells by reducing oxidative stress.METHODS Left ventricular of cardiac microvascular endothelial cells(CMECs),isolated from 8-week-old male C57BL/6N mice,were randomized into four groups:Control group,Vehicle group,H/R group,(H/R+Crocin group).The superoxide anion of CMECs,cell proliferation ability,cell migration ability and expression of Nox2 were measured respectively.RESULTS(1)Compared with Control group,H/R group had lower cell proliferation(P<0.05),lower cell migration ability and higher superoxide anion level(P<0.01).(2)Compared with H/R group,crocin increased cell proliferation,enhanced angiogenesis and decreased superoxide anion level(P<0.05).(3)Western Blot showed that the level of Nox2 in H/R group was higher than that in Control group(P<0.01).And after crocin pretreatment,the level of Nox2 was lower than that in H/R group(P<0.05).CONCLUSION These results suggest that Crocin protects CMECs against H/R injury through reducing the expression of Nox2 that decreases oxidative stress.
