摘要
Et试剂是由胆碱酯酶和DTNB〔5,5'-二硫代双(2-硝基苯甲酸)〕复合而成的一种农药残留快速检测专用试剂。在农药残留检测时直接将其与样品待测液进行酶抑制反应,使得在进行酶抑制反应过程时仅需三种试剂即可完成。检测方法更加简便,并减少实验器材的使用和环境的污染。Et试剂在(0~5℃)的温度下的保存时间大于510天。同时Et试剂具有良好的高温稳定性和低温抗冻融性,在室温保存65天以及在低温反复冻融五次后酶活性没有明显降低。将Et试剂应用于茶叶和果蔬中的有机磷和氨基甲酸酯类农药残留快速检测得到较好的结果。研发的高灵敏度Et试剂不仅能应用于茶叶、蔬菜中农药残留的快速检测而且也可拓展应用于水和土壤等其他样品中的农药残留检测。
Et reagent is a special reagent for rapid detection of pesticide residues,which is composed of cholinesterase and DTNB(5,5'-Dithiobis-(2-nitrobenzoic acid)).During the detection of pesticide residue,the enzyme inhibition reaction is carried out directly with the sample to be tested,so that only three reagents are needed to complete the enzyme inhibition re?action.The detection method is more convenient,and reduces the use of experimental equipment and environmental pollu?tion.The retention time of Et reagent at the temperature of(0~5℃)was more than 510 days.At the same time,Et reagent has good high temperature stability and low temperature freeze and thaw resistance,and the enzyme activity is not signifi?cantly reduced after 65 days of storage at room temperature and five times of repeated freeze and thaw at low temperature.Et reagent was applied to the rapid detection of organophosphorus and carbamate pesticide residues in tea leaves and fruits and vegetables.The developed high-sensitivity Et reagent can not only be applied to the rapid detection of pesticide residues in tea and vegetables,but also be extended to the detection of pesticide residues in other samples such as water and soil.
