摘要
解淀粉芽孢杆菌AF1能产生高活性淀粉酶、蛋白酶以及抗菌等活性物质。选取1月龄脱瘟土鸡苗120羽,进行随机分组,常规饲喂40 d。结果发现:AF1益生菌组和优菌肽组平均日增体质量显著高于空白对照组(P<0.05),而料肉比显著低于空白对照组(P<0.05)。刮取鸡盲肠内壁物检测病原菌,AF1益生菌组和优菌肽组的盲肠内大肠杆菌数明显比空白对照组少(P<0.01),且未检出沙门氏菌。采用RAPD技术筛选AF1菌株的特异性引物并进行PCR扩增,AF1益生菌组3只鸡肠道菌群的DNA条带相似度达到90%以上,与纯菌株AF1的条带有60%以上相同,空白对照组3只鸡的带谱也几乎完全相同,且2组之间的条带存在极显著差异。研究表明:AF1菌在饲喂土鸡时很可能定植在鸡肠道内影响肠道菌群,抑制病原菌生长,减少疾病发生,并能显著提高鸡饲料转化率,降低饲料成本。
High activities of amylase,proteinase and antibacteria from Bacillus amyloliquefaciens AF1 were detected.One hundred and twenty chickens with one month’s age were divided into 3 groups randomly:control group,AF1 group and youjuntai group.The chicken’s vitality,appetite,behavier,diarrhoea,livability and the amount of rest and feeding fodder were observed every day during a 40-day interval.Results showed that chicks in AF1 group and Youjuntai group were more active than the control group.Especially,AF1 group had a livability of 100%.ADG in AF1 group and Youjuntai group were significantly higher than that of the control group(P<0.05)and the F/G were significantly lesser than that of the control group(P<0.05).Quantification of Escherichia Coli in the chicken’s cecum showed that the pathogenic bacteria in AF1 group and youjuntai group was significantly decreased compared to the control group(P<0.01).Also,no Salmonella was detected in the chicken’s cecum of the AF1 group and Youjuntai group.Three AF1’s specific primers for PCR were screened by RAPD.It was showed that three chickens’intestinal flora in AF1 had a 90%in band similarity,with more than 60%similarity in bands to that of the single strain AF1.The band spectrums of the three chicken’s intestinal flora in control group were almost identical.The bands between AF1 group and the control group had highly significant difference.All above,AF1 can probably live in the chicken’s cecum and affect the intestinal flora by inhibiting the growth of pathogenic bacteria,which could reduce the disease and enhance the immunity and the survival rate.Accordingly,these beneficial of AF1 could significantly improve the feed conversion ratio and reduce the feed cost.
