摘要
目的探索二甲双胍(Metformin)对乳腺癌MCF7细胞的影响.方法荧光素酶报告实验检测miR-26a-5p和高迁移率族蛋白A2(high mobility group A2,HMGA2)靶向关系;蛋白印迹检测HMGA2蛋白表达;EdU染色检测细胞增殖;划痕实验检测细胞迁移;二甲双胍处理后,CCK8(cell counting Kit-8)法检测细胞存活率.划痕实验检测细胞迁移;实时定量PCR和蛋白印迹检测miR-26a-5p和HMGA2表达.结果miR-26a-5p可靶向HMGA2,过表达miR-26a-5p降低MCF细胞中HMGA2蛋白水平、增殖能力,减弱迁移能力,而通过二甲双胍处理后,miR-26a-5p表达水平增加,HMGA2蛋白水平、划痕愈合率降低.结论miR-26a-5p靶向HMGA2减弱乳腺癌MCF7细胞增殖和迁移能力.
ObjectiveTo explore the effect of metformin on MCF7 breast cancer cells.Methods Luciferase assay was used to detect the targeting relationship between miR-26a-5p and high mobility group A2(HMGA2).HMGA2 protein expression was detected by Western blot-ting,cell proliferation by EdU staining and cell migration by scratch test.After metformin treatment,the cell survival rate was determined by cell counting kit-8(CCK8)method.Cell migration was de-tected by scratch test.The expression levels of miR-26a-5p and HMGA2 were measured by real-time quantitative PCR and Western blotting.Results miR-26a-5p could target HMGA2,and overex-pression of miR-26a-5p could reduce HMGA2 protein level and proliferative and migrating ability of MCF cells.However,after treatment with metformin,the expression level of miR-26a-5p was in-creased,and the HMGA2 protein level and scratch healing rate were decreased.Conclusion miR-26a-5p attenuates the proliferation and migration of MCF7 breast cancer cells by targeting HMGA2.
