小儿伤科方泡腾颗粒和配方颗粒对闭合性骨折大鼠的治疗效果研究 被引量:5

Effectiveness of Pediatric Traumatology Recipe Effervescent Granules and Formula Granules in Treatment of Rats with Closed Fracture

摘要 目的探讨小儿伤科方泡腾颗粒和配方颗粒对闭合性骨折大鼠的治疗效果。方法将100只SD大鼠随机分为正常组、骨折模型组、传统汤液组、泡腾颗粒组以及配方颗粒组,每组20只。除正常组外其余各组均制作闭合性骨折大鼠模型,传统汤液组、泡腾颗粒组以及配方颗粒组均给予小儿伤科方按生药量计算10 g/kg灌胃,正常组及骨折模型组给予等量生理盐水灌胃。治疗30 d后检测各组的全血黏度、血浆黏度、血管内皮生长因子(VEGF)、碱性磷酸酶(ALP)、D-二聚体、血清钙、血清磷浓度,根据抗折力实验以及影像学结果判定愈合组织抗折强度和X线片评分,并对骨折组织进行HE染色观察。结果与骨折模型组比较,传统汤液组、泡腾颗粒组和配方颗粒组的全血黏度、血浆黏度、VEGF、ALP、D-二聚体、血清磷浓度均显著降低(P<0.05),血清钙浓度、骨折愈合抗折强度及X线片评分均显著升高(P<0.05);但3用药组之间上述指标比较差异均无统计学意义(P>0.05)。骨折模型组骨折部位可见明显的组织分离,小儿伤科方治疗后骨折部位出现新生的骨小梁,可见较成熟的骨组织,并且局部形成骨连接。结论小儿伤科方泡腾颗粒和配方颗粒能通过改善血液循环,促进骨小梁、骨组织的生长,从而调节骨组织修复及再生,促进大鼠骨折愈合。 Objective To investigate effectiveness of pediatric traumatology recipe Effervescent Granules and Formula Granules in treatment of rats with closed fracture.Methods A total of 100 SD rats were randomly divided into normal group,fracture model group,traditional decoctions group,effervescent granule group and formula granule group(n=20 in each group).Closed fracture rat models were made in all groups except for normal group.Traditional decoctions,effervescent granule and formula granule groups were lavaged with 10 g/kg pediatric traumatology recipe(crude drug),while normal and fracture model groups were lavaged with the same volume of physiological saline.After treatment for 30 d,levels of whole blood viscosity,plasma viscosity,vascular endothelial growth factor(VEGF),alkaline phosphatase(ALP),D-dimer,serum calcium and serum phosphorus concentration were detected according to the flexural strength test.Rupture strength and X-ray film score of healing tissues were judged according to rupture ability test and iconography results,and fracture tissues were observed by hematoxylin and eosin(HE)staining.Results Compared with those in fracture model group,levels of whole blood viscosity,plasma viscosity,VEGF,ALP,D-dimer and serum phosphorus concentration were significantly decreased(P<0.05),while values of serum calcium concentration,fracture healing rupture strength and X-ray film scores were significantly increased in traditional decoctions,effervescent granule and formula granule groups(P<0.05),but there were no significant differences in the above indexes among three administration groups(P>0.05).In fracture model group,obvious tissue separation was observed in fracture site.After treatment of pediatric trauma recipe,new bone trabeculae appeared in the fracture site,and more mature bone tissues were observed,and local synostosis was formed.Conclusion Pediatric traumatology recipe Effervescent Granules and Formula Granules may regulate repair and regeneration of bone tissues and promote fracture healing by improving blood circulation and growth of bone trabecula and bone tissues of rats.
