摘要
目的:探讨大黄素对大鼠烫伤创面模型皮肤表皮细胞增殖的作用。方法:选取健康SD大鼠120只,雌雄各半,随机分为对照组、模型组、湿润烧伤膏组、大黄素低浓度组及大黄素高浓度组,各24只。模型组、湿润烧伤膏组、大黄素低浓度组及大黄素高浓度组大鼠采用80℃水浴法建立深Ⅱ度烫伤创面模型,对照组大鼠给予25℃水浴。模型建立后,模型组和对照组给予生理盐水外擦,1次/d;湿润烧伤膏组给予湿润烧伤膏1 g外敷,1次/d;大黄素低浓度组给予150μg/g大黄素软膏1 g外敷,1次/d;大黄素高浓度组给予200μg/g大黄素软膏1 g外敷,1次/d;各组大鼠连续用药21 d。对大鼠创面愈合时间、创面愈合率、创面皮肤组织毛细血管数、细胞角蛋白19(K19)阳性细胞数、表皮生长因子(EGF)、血管内皮生长因子(VEGF)水平进行检测。结果:治疗后,与模型组比较,各组大鼠创面愈合时间较短,且大黄素低浓度组>大黄素高浓度组、湿润烧伤膏组,差异有统计学意义(P<0.05)。给药后7、14、21 d,与模型组比较,各组大鼠创面愈合率、EGF水平、VEGF水平较高,皮肤毛细血管数、K19阳性细胞数较多,且大黄素低浓度组<大黄素高浓度组、湿润烧伤膏组,差异有统计学意义(P<0.05)。结论:大黄素能加速大鼠烫伤模型创面愈合,且药物浓度越高效果越明显,其作用可能与上调EGF、VEGF表达,促进皮肤表皮细胞增殖及毛细血管生成有关。
Objective:To analyze the effect of emodin on the epidermal cells proliferation of wound skin in scalded rats.Methods:A total of 120 healthy SD rats,half male and female,were selectedand randomly divided into control group,model group,moist burn ointment group,emodin low-concentration group and emodin high-concentration group,with 24 rats in each.Rats in model group,moist burn ointment group,emodin low-concentration group and emodin high-concentration group were established deep second degree scald wound models by 80℃water bath method and rats in the control group received 25℃water bath.After the model was established,the model group and the control group were given normal saline external rubbing,once a day.The moist burn ointment group was given moist burn ointment 1 g for external application,once a day.The emodin low-concentration group was given 150μg/g emodin ointment 1 g for external application,,once a day.The emodin high-concentration group was given 200μg/g emodin ointment 1 g for external application,once a day.The wound healing time,wound healing rate,the numbers of capillaries,cytokeratin 19(K19)positive cells,epidermal growth factor(EGF)and vascular endothelial growth factor(VEGF)levels in skin tissue of wound surface were detected.Results:After treatment,compared with the model group,the wound healing time was shorter in each group,and emodin low-concentration group’s healing time>emodin high-concentration group and moist burn ointment group’s healing time and the differences were statistically significant(P<0.05).After 7,14,21 d administration,compared with the model group,the wound healing rate,EGF levels and VEGF levels were higher,the number of capillaries in the skin and the number of K19 positive cells were more in each group,and emodin low-concentration group’s<emodin high-concentration group and moist bum ointment group1 s and the difference were statistically significant(P<0.05).Conclusions:Emodin can accelerate wound healing in scalded r a ts,and the higher the drug concentration is,the more obvious the effect is.The effect may be related to up-regulating EGF and VEGF expressions,promoting skin epiderm al cell proliferation and capillary production.
