摘要
目的:探讨黄芩素促血管舒张的作用及作用机制。方法:黄芩素孵育大鼠心脏微血管内皮细胞24 h,MTT法检测细胞活力;硝酸还原酶法检测细胞上清NO含量;ELISA法检测细胞e NOS蛋白表达。黄芩素孵育大鼠胸主动脉环24 h,去甲肾上腺素(10^-6mol/L)收缩血管后应用乙酰胆碱(10^-8,10^-7,10^-6,10^-5mol/L)舒张血管,检测血管张力变化。结果:黄芩素(0.5,5,20μm)具有促进大鼠心脏微血管内皮细胞NO产生与e NOS蛋白表达的作用。一氧化氮合酶抑制剂L-NAME(100μm)能够抑制黄芩素促NO产生的作用。黄芩素(20μm)具有增强乙酰胆碱舒张动脉环的作用。结论:黄芩素可能通过促进内皮细胞e NOS蛋白表达与NO释放这一途径增强乙酰胆碱对血管的舒张作用。
Objective:To study the chronic effect of baicalein on Ach-induced vasorelaxation and its mechanism.Methods:Rat cardiac microvascular endothelial cells were incubated with baicalein for 24 h.Cell viability was detected by MTT.NO was detected by nitrate reductase.The e NOS protein expression was detected by ELISA.Rat thoracic aorta was incubated with baicalein for24 h,which was contracted by NE(10^-6 mol/L)and relaxed by Ach(10^-8,10^-7,10^-6,10^-5 mol/L).Vascular tension was detected by tension transducer.Results:Baicalein(0.5,5,20μm)increased NO production which was suppressed by L-NAME(100μm)and e NOS protein expression in rat cardiac microvascular endothelial cells.Baicalein(20μm)reduced Ach-induced vasorelaxation in rat thoracic aorta.Conclusion:Baicalein might reduce Ach-induced vasorelaxation via increasing e NOS expression and NO production in endothelial cells.
