摘要
目的观察温肾固疏方对绝经后骨质疏松症肾阳虚型患者类固醇激素受体辅激活子3(SRC-3)、转录元件辅激活蛋白(PGC-1a)、B细胞淋巴瘤基因-2(Bcl-2)的蛋白表达及骨髓间充质干细胞Notch信号通路的影响。方法将86例绝经后骨质疏松症肾阳虚型患者按照随机数字表法分为2组。对照组43例予西医常规治疗;治疗组43例在对照组治疗基础上加用温肾固疏方治疗。2组均治疗8周。记录2组治疗前后骨密度(BMD)变化;比较2组治疗前后SRC-3、PGC-1a、Bcl-2的表达;比较2组治疗后Notch通路表达情况;比较2组疗效。结果 2组治疗后BMD均较本组治疗前增加(P<0.05),且治疗组增加更明显(P<0.05)。2组治疗后SRC-3、PGC-1a、Bcl-2均较本组治疗前升高(P<0.05),且治疗组升高更明显(P<0.05)。治疗组治疗后Notch通路关键分子(Notch1、Jagged1、Hes1)mRNA表达均高于对照组(P<0.05)。治疗组总有效率79.07%,对照组总有效率62.79%,治疗组疗效优于对照组(P<0.05)。结论温肾固疏方治疗绝经后骨质疏松症肾阳虚型,能提高患者BMD,上调SRC-3、PGC-1a、Bcl-2的蛋白表达水平,恢复骨髓间充质干细胞功能。
Objective To observe the effect of Wenshen Gushu recipe on the expression of steroid receptor coactivator 3(SRC-3), transcription element coactivator protein(PGC-1 a), B-cell lymphoma gene-2(Bcl-2)and the Notch signaling pathway of bone marrow mesenchymal stem cells in postmenopausal osteoporosis patients with kidney yang deficiency type. Methods 86 postmenopausal osteoporosis patients with kidney yang deficiency type were divided into two groups according to random number table method. 43 cases in control group were treated by conventional western medicine. 43 cases in treatment group were treated by Wenshen Gushu recipe on the basis of the control group. Both groups were treated for 8 weeks. The changes of bone mineral density(BMD) before and after treatment were recorded in the two groups. The protein expression of SRC-3, PGC-1 a and Bcl-2 before and after treatment in the two groups were compared. The expression of Notch pathway after treatment in the two groups was compared. The curative effect of the two groups was compared. Results The BMD in both groups was increased after treatment(P<0.05), and the increase in the treatment group was more significant(P<0.05). After treatment, SRC-3, PGC-1 a and Bcl-2 in the two groups were higher than those in the group before treatment(P<0.05), and the increase in the treatment group was more significant(P<0.05). The mRNA expression of Notch pathway key molecules(Notch 1, Jagged1, Hes1)in the treatment group was higher than that in the control group(P<0.05). The total effective rate was 79.07% in treatment group and 62.79% in control group. The clinical curative effect of the treatment group was better than that of the control group(P<0.05). Conclusion Wenshen Gushu recipe can improve BMD, up-regulate protein expression of SRC-3, PGC-1 a and Bcl-2 in postmenopausal osteoporosis patients with kidney yang deficiency type, and restore the function of bone marrow mesenchymal stem cells.
