摘要
目的:对光石韦的急性毒性、对凝血作用的影响和止咳作用进行研究。方法:采用灌胃给药方式观察光石韦对小鼠的急性毒性反应;对正常大鼠予光石韦水提物进行灌胃给药,测定大鼠凝血酶原时间(PT)、凝血酶(TT)、活化部分凝血活酶时间(APTT),观察光石韦对大鼠凝血功能的影响;对正常小鼠用光石韦水提物进行灌胃,用引咳仪对小鼠进行二氧化硫引咳,测定咳嗽潜伏期和60、90 s咳嗽次数,观察光石韦水提物对小鼠止咳作用。结果:光石韦急性毒性的最大耐受量为118.0 g·kg-1,14 d后小鼠无死亡;光石韦提取物组高、低剂量组大鼠PT显著增加(P<0.01),中、低剂量组大鼠APTT值显著增加(P<0.01);光石韦提取物组高剂量组小鼠90s咳嗽次数显著减少(P<0.05)。结论:光石韦经口给药急性毒性低,可改善大鼠凝血功能,具有一定的止咳作用。
Objective:To study the acute toxicity of Pyrrosiae calvatae folium and its effect on the blood coagulation function and antitussive effect.Methods:The acute toxicity was measured in normal mice,which were administered intragastrically with aqueous extract of Pyrrosiae calvatae folium;the serum prothrombin time(PT),thrombin time(TT),activated partial thromboplastin time(APTT)were measured in normal rat,which were administered intragastrically with different doses of aqueous extract of yrrosiae calvatae folium in order to investigate the effect on blood coagulation function of Pyrrosiae calvatae folium;the latent period of coughing,coughing times in 60s,coughing times in 90s were measured in normal mice which were induced coughing by sulfur dioxide,the mice were administered different doses of aqueous extract of Pyrrosiae calvatae folium in order to investigate the antitussive effect on mice.Results:The maximal tolerated dose of Pyrrosiae calvatae folium was 118.0 g/kg·bw on the mice oral acute toxicity test,14 d after the administration no mouse was dead;PT of low-dose and high-dose groups of Pyrrosiae calvatae folium increase significantly(P<0.01);the APTT of low-dose and middle-dose groups of Pyrrosiae calvatae folium increased significantly(P<0.01);the coughing times in 90s of high-dose group of Pyrrosiae calvatae folium decreased significantly(P<0.05).Conclusion:Pyrrosiae calvatae folium was classified as low-toxic material,Pyrrosiae calvatae folium has an improving effects on blood coagulation function and a certain antitussive effect.
