摘要
目的研究草果、菱角壳水提混合物对急性酒精中毒后小鼠解酒效用及对酒精性肝损伤的保护功效。方法将小鼠随机分为5组,清醉组,草果菱角壳水提混合液高(1.0 g/mL)、低(0.5 g/mL)剂量组,空白组和模型组。各给药组予相应药物,给药后30 min,空白组予蒸馏水,余下4组按0.015 m L/g灌胃56°白酒,建立急性酒精中毒模型,记录小鼠醉酒与醒酒时长。另取小鼠,按同样方法分组并给药7 d,建造急性肝损伤模型,末次白酒灌胃3 h后,采血取肝脏,测定肝指数,用相应测试盒测定血清和肝匀浆相关生化指标。结果相较空白组,模型组乙醇脱氢酶(ADH)、总超氧化物歧化酶(SOD)活性降低(均P<0.01),肝指数、谷丙转氨酶(ALT)、谷草转氨酶(GOT)、丙二醛(MDA)均增高(P<0.05或P<0.01);相较模型组,清醉组、高剂量组醉酒时长延缓(P<0.01),醒酒时长缩减(P<0.05),清醉组、低剂量组ADH升高(P<0.05),清醉组、高、低剂量组肝指数、GOT、ALT、MDA大幅下降(P<0.05或P<0.01),SOD显著升高(均P<0.01)。结论草果、菱角壳水提混合液对急性酒精性中毒小鼠起解酒效用,并在一定程度上保护肝脏免受急性酒精性损伤,其机制推测与脂质对氧化应激的抵抗能力、O2-清除和加速酒精排泄有关。
Objective:To investigate the antialcoholism effect of the water extract mixture of Amomum tsao-ko on acute alcoholism mice and protection effect on acute alcoholic liver injury.Methods:50 mice were randomly divided into five groups:Qingzui group,the water extract mixture of Amomum tsao-ko and hull of water chestnut high dose group(1.0 g/mL)and low dose group(0.5 g/mL),the blank group and the model group.30 minutes after administration,distilled water was given to the blank group,and 0.015 mL/g 56 liquor was given to the remaining four groups.The acute alcoholism model was established and the duration of intoxication and sobering up was recorded.The same method was used for 7 d to establish acute liver injury models.Three hours after the last infusion of liquor,blood and liver were collected;liver index was measured,and serum and liver homogenate related biochemical indexes were measured with corresponding test boxes.Results:Compared with the blank group,the activity of ADH and SOD were decreased(P<0.01),liver index,ALT,GOT and MDA were increased(P<0.05 or P<0.01).Compared with the model group,the duration of intoxication was delayed(P<0.01);the duration of sobering up was prolonged in Qingzui group and high dose group(P<0.05);ADH in Qingzui group and low dose group was increased(P<0.05);liver index,ALT,GOT and MDA in Qingzui group,high and low dose group were significantly decreased(P<0.05 or P<0.01);SOD was significantly increased(P<0.01).Conclusion:The water extract mixture of Amomum tsao-ko and hull of water chestnut has the effect of dissolving alcohol and protecting liver from acute alcohol injury to some extent.The mechanism is speculated to be related to the resistance of lipid to oxidative stress,O2 clearance and acceleration of alcohol excretion.
