摘要
目的探讨Sema4D通过调节趋化因子和CD8^+T细胞募集促进口腔扁平胎癣的发展机制。方法回顾性选择宝鸡市人民医院收集的口腔扁平胎癣病理标本34例作为病例组,同期选择正常口腔黏膜组织21例作为对照组。采用免疫组化法检测两组组织CXC类趋化因子受体7(CXCR7)与Sema4D表达情况,采用流式细胞仪检测两组血液CD8^+T细胞,并分析病例组CXCR7、Sema4D表达阳性率与CD8^+T细胞含量的相关性。结果病例组组织的CXCR7与Sema4D表达阳性率分别为82. 4%、88. 2%,显著高于对照组组织的33. 3%和42. 9%(P <0. 05)。病例组的CD8^+T细胞含量显著高于对照组,CD4^+T细胞含量显著低于对照组(P <0. 05)。直线相关分析显示口腔扁平苔藓患者病理组织的CXCR7、Sema4D表达阳性率与CD8^+T细胞含量成显著正相关性,与CD4^+T细胞含量呈显著负相关性(P <0. 05)。COX回归分析显示CXCR7、Sema4D表达阳性率均为影响CD8+T含量的主要因素(P <0. 05)。结论 Sema4D可通过调节趋化因子CXCR7和CD8^+T细胞募集促进口腔扁平胎癣的发展,是监测T细胞免疫功能的关键标志分子,可能在口腔扁平胎癣的发生中发挥重要作用。
Objective To investigate the mechanism that Sema4 D promotes the development of oral lichen planus by regulating chemokines and the recruitment of CD8^+T cells. Methods A total of 34 cases of oral lichen planus collected from People’s Hospital of Baoji City retrospectively selected as the case group. 21 cases of normal oral mucosa were selected as the control group. The expression of CXCR7 and Sema4 D in the two groups were detected by immunohistochemistry. The blood CD8^+T cells were detected by flow cytometry. And the correlations between the positive rate of CXCR7 and Sema4 D and the content of CD8^+T cells in the case group were analyzed. Results The positive rates of CXCR7 and Sema4 D in the case group were 82. 4% and 88. 2%,respectively,which were significantly higher than those in the control group( 33. 3% and42. 9%)( P < 0. 05). The content of CD8^+T cell in the case group were significantly higher than that in the control group,and the content of CD4^+T cell were significantly lower than that in the control group( P < 0. 05). Linear correlation analysis showed that the positive rates of CXCR7 and Sema4 D in pathological tissues of patients with oral lichen planus were significantly positively correlated with CD8^+T cell content,and significantly negatively correlated with CD4^+T cell content( P < 0. 05). COX regression analysis showed that the positive rates of CXCR7 and Sema4 D were the main factors affected CD8^+T content( P < 0. 05). Conclusion Sema4 D can promote the development of oral lichen planus by regulating chemokines CXCR7 and recruiting CD8^+T cells. It is a key marker for monitoring the immune function of T cells and may play an important role in the occurrence of oral lichen planus.
