摘要
目的了解北京地区犬钩端螺旋体的感染情况。方法采用快速实时荧光快速聚合酶链式反应(Rapid real-time fluorescence quantitative polymerase chain reaction, qPCR)对2017年1月至2019年2月从宠物医院、犬舍、流浪动物基地收集到的1 600份血液和尿液样本进行分子学检测,以及风险因素分析。并对qPCR阳性病例样本进行血清学和PCR鉴定。结果 qPCR检出阳性样本17份,检出率为1.01%(95%CI 0.6-1.7)。其中尿液阳性检出率2.00%(16/800)显著高于血液检出率0.13%(1/800)(χ~2=11.653,P<0.01)。地区(χ~2=17.828,P<0.05)、季节(χ~2=10.916,P<0.05)、样本来源(χ~2=9.432,P<0.01)是犬钩体感染的风险因素,但不同年龄(χ~2=1.103,P=0.613)、性别(χ~2=1.124,P=0.223)、品种(χ~2=0.427,P=0.448)的犬感染钩体的差异无统计学意义。显微凝集试验显示12份qPCR阳性病犬血清样本存在澳洲群和拜伦群抗体;菌株经PCR鉴定均为问号钩端螺旋体。结论北京部分地区存在犬钩体病的流行,应加强疾病监测和宠物犬定期疫苗接种以降低其向人类传播的风险。
The aim of this study was to investigate the prevalence of canine leptospirosis in Beijing. Blood samples and urine samples(n=1 600) were collected from pet hospitals, kennels and shelters during January 2017 to February 2019. All samples were performed by quantitative real-time polymerase chain reaction(qPCR) assays. Then antibody detection and conventional PCR assays were conducted for qPCR-positive samples. The overall positive rate of qPCR was 1.01%(17/1 600). While the positive rate of urine samples(2.00%) was significant higher than that of blood samples(0.13%). The chi-square test showed region(χ~2=17.828, P<0.05) season(χ~2=10.916, P<0.05) and source(χ~2=9.432, P<0.01) were major risk factors associated with leptospirosis. However, no significant differences were found among age(χ~2=1.103, P=0.613), gender(χ~2=1.124, P=0.223) and breed(χ~2=0.427, P=0.448) in infected dogs. Microscopic agglutination test(MAT) results showed 12 serum samples were positive for the antibodies of Australia serogroup and Ballum serogroup. The isolates were identified as Leptospira interrogans by PCR. This study indicates the canine leptospirosis prevalence in part of Beijing. Disease surveillance and vaccination at a regular interval should be recommended for dog owners to reduce the risk of infection and transmission to human.
