摘要
通过单因素试验和正交试验确定超声波辅助条件下有机溶剂/盐双水相体系对桑葚色素的最佳萃取条件,并考察桑葚色素的体外抗氧化性。结果表明, pH为4.0左右乙醇与硫酸铵形成的双水相体系稳定,超声波辅助双水相萃取桑葚色素最佳工艺为硫酸铵质量分数16%、乙醇体积分数25%、超声功率300 W、超声时间30 min。此条件下桑葚色素回收率可达80.5%。桑葚色素对羟自由基(·OH)和超氧阴离子(O2^-·)的清除率高于BHT、PG和VC,对1, 1-二苯基-2-苦苯肼自由基(DPPH·)清除率高于BHT、PG,但略低于VC。
The aqueous two-phase extraction conditions for ultrasonic-assisted of pigment from mulberry is studied by applying single factor and orthogonal experimental designs. Its antioxidation in vitro is assayed. The experimental results show that the two-phase system formed by ethanol and ammonium sulfate is stable with pH 4.0. The optimal ultrasoundassisted aqueous two-phase extraction conditions of pigment from mulberry are ammonium sulfate concentration of 16%, ethanol concentration of 25%, ultrasonic power of 300 W, and ultrasonic time of 30 min. The yield of pigment from mulberry reaches 80.5% by this system. The scavenging capacities of pigment from mulberry on superoxide anion radical and hydroxyl free radical are more efficient than BHT, PG and VC. The scavenging capacity of pigment from mulberry on 1, 1-diphenyl 2-picrylhydrazyl is stronger than BHT and PG, but weaker than that of VC.
