摘要
为提高屎肠球菌B13乳酸菌素的产量,以接种量、培养基初始pH值和发酵温度为考察因素,在单因素试验的基础上,利用Box-Behnken中心组和试验设计原理进行三因素三水平的响应面优化分析,优化后的最佳培养条件为:接种量4.4%、培养基初始pH 5.3、发酵温度33℃。在此条件下,乳酸菌素的抑菌效价达5.32×10^8 IU/mL,比优化前提高1.44倍,为屎肠球菌B13乳酸菌素的开发利用提供参考依据。
In order to improve the bacteriocin production of Enterococcus faecium B13,inoculum,initial pH of the medium and fermentation temperature was taken as an investigation factor,on the basis of single factor experiment,four factors were designed using Design Expert 8.0.6 Box-Behnken center composite design. The optimal fermentation conditions were inoculum amount of 4.4 %,initial pH of 5.3,fermentation temperature of33 ℃. Under this condition,the bacteriocin inhibitory activity was 5.32 ×10^8 IU/mL,which was 1.44 times higher than before,it provides a reference for the development and utilization of Enterococcus faecium B13 lactobacillus.
