摘要
目的:建立紫萍与青萍中黄酮类成分测定的方法,对其含量测定结果进行多变量统计分析。方法:采用高效液相色谱四级杆飞行时间串联质谱(HPLC-QTOF-MS)法,色谱柱:Agilent ZORBAX Extend-C18RRHT(2.1 mm×50 mm,1.8μm);以甲醇为流动相A,以0.1%甲酸溶液为流动相B,梯度洗脱;流速为0.3 mL·min-1;柱温为30℃;进样量为1μL;测定52批浮萍样品中荭草苷、异荭草苷、牡荆苷、木犀草苷、木犀草素、芹菜素6种黄酮类成分含量,并采用主成分分析(PCA)和正交偏最小二乘法-判别分析(OPLS-DA)进行多变量统计分析。结果:6种黄酮类化合物的精密度、重复性和稳定性良好(RSD≤4.0%),平均加样回收率在95.41%~105.82%,RSD为2.03%~4.16%(n=6)。HPLC-QTOF-MS法结合主成分聚类分析宜用于比较不同品种浮萍中黄酮类成分含量差异。结论:采用PCA与OPLS-DA进行多维变量统计分析与测定结果相符,HPLC-QTOF-MS结合化学计量学方法能够评价不同浮萍中的6种黄酮类化合物。
Objective: The study was performed to develop a qualitative method to determine six flavonoids in Spirodela polyrrhiza(L.) Schleid and Lemna minor L.. The study included the application of multivariate statistical analysis to the testing results. Methods: An HPLC-QTOF-MS method with an Agilent ZORBAX Extend-C18 RRHT column(2.1 mm × 50 mm, 1.8 μm) was applied. The injection volume was 1 μL at the flow rate of 0.3 mL·min-1. The column temperature was 30 ℃. The mobile phase was composed of methanol(A)-water(B) for gradient elution. Six flavonoids in 52 duckweed samples were measured, including orientin,isoorientin, vitexin, galuteolin, luteolin and apigenin. PCA(principal components analysis) and OPLS-DA(orthoganol partial least squares-discriminant analysis) were used for multivariate statistical analysis. Results:The method for determination of six flavonoids in the study showed high precision, repeatability and stability(RSD≤4.0%);the average recoveries were within the range of 95.41%-105.82%, and the RSDs were within the range of 2.03%-4.16%(n=6). The method of HPLC-QTOF-MS combined with PCA can be used to identify the contents of flavonoids in different duckweed species. Conclusion: Results from PCA and OPLS-DA applied in multivariate statistical analysis confirmed those of manual identification. HPLC-QTOF-MS combined with chemometrics analysis was applicable for the evaluation of 6 flavonoids in two duckweed species.
