摘要
为了检测不同提取方法获得的浒苔提取物浒苔多糖1、浒苔多糖2、浒苔多糖3和浒苔多糖硒的体外抗禽流感病毒活性,试验将4种浒苔多糖用细胞维持液倍比稀释,以金刚烷胺、病毒灵、黄芪多糖和紫锥菊提取物为对照,以犬肾细胞(MDCK)为细胞模型,观察细胞病变效应(CPE),并采用MTT法研究4种浒苔多糖对H9N2禽流感病毒的直接杀灭、抑制病毒复制和阻断病毒吸附的治疗指数。结果表明:浒苔多糖1和浒苔多糖2直接杀灭H9N2禽流感病毒作用治疗指数分别为7.33和7.29,该指数低于金刚烷胺(33.56)、病毒灵(36.00)和黄芪多糖(15.78),高于紫锥菊提取物(6.92);浒苔多糖1和浒苔多糖2抑制H9N2禽流感病毒复制作用的治疗指数分别为5.78和6.13,该指数低于金刚烷胺(22.68)、病毒灵(26.71)和黄芪多糖(10.18),高于紫锥菊提取物(5.41);浒苔多糖1和浒苔多糖2阻断H9N2禽流感病毒吸附作用的治疗指数分别为13.83和12.75,该指数低于金刚烷胺(46.61)、病毒灵(55.20)和黄芪多糖(16.25),高于紫锥菊提取物(11.22);浒苔多糖3和浒苔多糖硒不具有直接杀灭病毒、抑制病毒复制和阻断病毒吸附的作用。说明浒苔多糖1和浒苔多糖2具有体外抗禽流感病毒活性,表现为对H9N2禽流感病毒具有直接杀灭、抑制病毒复制和阻断吸附的作用,浒苔多糖3和浒苔多糖硒无体外抗禽流感病毒活性。
In order to detect the anti-avian influenza virus activity of Brassica polysaccharide 1,Brassica polysaccharide 2,Brassica polysaccharide 3 and Brassica polysaccharide selenium obtained by different extraction methods in vitro.The four kinds of canola polysaccharides were diluted with cell maintenance solution,and amantadine,virion,astragalus polysaccharide and echinacea extract were used as controls,and the canine kidney cells(MDCK)were used as the cell model to observe the cytopathic effect(CPE).The MTT method was used to detect the therapeutic index of four kinds of Brassica polysaccharides for direct killing of H9 N2 avian influenza virus,inhibition of viral replication and blocking of virus adsorption.The results showed that the therapeutic index of direct application of Brassica polysaccharide 1 and Brassica polysaccharide 2 directly killing H9 N2 avian influenza virus was 7.33 and 7.29,respectively,which was lower than that of amantadine(33.56),virgin(36.00)and astragalus polysaccharide(15.78),higher than Echinacea extract(6.92);The therapeutic indices of the inhibition of H9 N2 avian influenza virus replication by Brassica polysaccharide 1 and Brassica polysaccharide 2 were 5.78 and 6.13,respectively,which was lower than that of amantadine(22.68),virgin(26.71)and astragalus polysaccharides(10.18),higher than Echinacea extract(5.41).The therapeutic indices of the adsorption of H9 N2 avian influenza virus by Brassica polysaccharide 1 and Brassica polysaccharide 2 were 13.83 and 12.75,respectively,which was lower than that of amantadine(46.61),virgin(55.20)and astragalus polysaccharide(16.25),higher than Echinacea extract(11.22);The therapeutic indices of direct killing H9 N2 avian influenza virus,inhibition of viral replication and blocking of virus adsorption of Brassica polysaccharide 3 and Brassica polysaccharide selenium were zero.It indicated that Brassica polysaccharide 1 and Brassica polysaccharide 2 had anti-avian influenza virus activity in vitro,which had the effect of directly killing,inhibiting virus replication and blocking adsorption of H9 N2 avian influenza virus.Brassica polysaccharide 3 and Brassica Polysaccharide selenium had no anti-avian influenza virus activity in vitro.
