高效液相色谱—串联质谱法同时检测猪肉中125种兽药残留 被引量:15

Simultaneous Determination of 125 Veterinary Drug Residues in Pork by High Performance Liquid Chromatography-Tandem Mass Spectrometry

摘要 本方法建立了猪肉中包括磺胺类、喹诺酮类、硝基咪唑类、氯霉素类、β-受体激动剂类、大环内酯类、苯并咪唑类、糖皮质激素类等在内的125种兽药的高效液相色谱—串联质谱(HPLC-MS/MS)同时检测方法。样品经使用WondaPak QuEChERS多兽药残留专用提取包和净化包快速前处理后,通过多反应监测模式检测。对空白样品添加浓度1.0,5.0和10.0μg·kg-1后,考察回收率和相对标准偏差(RSD),结果显示:1.0μg·kg-1加标浓度的平均回收率为42.06%~121.14%,RSD为2.36%~19.07%;5.0μg·kg-1加标浓度的平均回收率为55.10%~112.84%,RSD为2.50%~19.65%;10.0μg·kg-1加标浓度的平均回收率为56.44%~114.57%,RSD为1.08%~18.45%。结果表明该方法具有前处理简便、测定时间短、通量大、重现性和准确度好等优点。 A high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)method was developed for the simultaneous quantification of 125 veterinary drugs in pork including sulfanilamides,quinolones,nitroimidazoles,chloramphenicola,β-agonists,macrolides,benzimidazoles and glucocorticoids.The meat samples were extracted and purified with WondaPak QuEChERS kits designed for multi-veterinary drug preparation,and then quantified by HPLC-MS/MS in multiple reaction monitoring mode.The method accuracy was investigated by calculating the recovery rates by adding standard mixture into blank sample at different concentrations of 1.0,5.0 and 10.0μg·kg-1,respectively.The experiments were repeated three times to evaluate the method reproducibility.The recovery rates of different adding concentrations of 1.0,5.0 and 10.0μg·kg-1 were 42.06%~121.14%,55.10%~112.84%and 56.44%~114.57%,respectively.Correspondingly,the relative standard deviation(RSD)of replications were 2.36%~19.07%,2.50%~19.65%and 1.08%~18.45%,respectively.The method was indicated to have many merits,including easy sample preparation,short analysis time,high reproducibility and accuracy,which can be used for high-throughput analysis of veterinary in meat samples.
