摘要
过氧化物酶体增殖物激活受体γ(PPARγ)是体内脂肪形成的必需转录因子,处于脂质沉积过程中信号传递通路的核心枢纽位置,促进多能间充质干细胞向脂肪细胞的定向、增殖分化和脂质沉积。本文首先介绍了PPARγ的基本结构和调控靶基因的基本特征,然后以脂肪细胞形成和脂肪沉积为主线,综述了PPARγ信号通路在脂肪细胞形成、脂肪沉积和脂肪因子分泌、营养水平等脂肪代谢过程的调控作用,最后对PPARγ与生物钟相互反馈调节关系以及其在脂质沉积中扮演重要作用进行了概述,对进一步深入综合了解PPARγ对脂肪代谢调控的分子机制具有重要意义,为脂肪代谢及相关疾病的人工干预提供靶基因和新途径。
Preoxisome proliferator-activated receptor gamma(PPARγ)is an essential transcription factor for fat formation in the body.It is located the central position of signal transmission which regulates the genes expression relating the lipid deposition,and it not only promotes the orientation of multidirectional mesenchymal stem cells to fat cells,but also promotes the proliferation,differentiation and lipid deposition of fat cells.First,the basic structure of PPARγ and the basic characteristics of regulatory target genes were introduced.Then,the regulatory effects of PPARγ on adipocyte formation,lipid deposition and fat factor secretion were reviewed according to the adipocyte formation and fat deposition as the main lines.Finally,the feedback regulation between PPARγand biological clock and its important role were instructed in lipid deposition,and high fat intake causes elevated transcription factor PPARγ to disrupt the normal biological clock.The review indicated that further understand the structure and function of PPARγ,the molecular mechanism of PPARγregulation of fat metabolism,and provides target genes and new approaches for artificial intervention of fat metabolism and related diseases.
