摘要
目的分析载脂蛋白E(APOE)及载脂蛋白A1(APOA1)基因与中老年人生化指标和简易精神状态量表(MMSE量表)评分的相关性。方法2019年1—6月对165例于湘潭市中心医院接受健康体检的中老年人进行MMSE量表评分调查,检测血清空腹血糖(FBG)、三酰甘油(TG)、胆固醇水平;提取基因组DNA,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)分析方法,对APOE、APOA1基因进行扩增和酶切,分析目标基因与生化指标和MMSE量表评分的相关性。结果APOE基因中,ε4携带者胆固醇水平高于非ε4携带者(P=0.017),MMSE量表评分低于非ε4携带者(P=0.005);APOA1+83 bp基因中,T携带者胆固醇水平低于非T携带者(P=0.032)。FBG、TG水平与各基因型无相关性(P>0.05)。结论APOE、APOA1+83 bp基因多态性与血清胆固醇水平存在相关性,APOE基因型与MMSE量表评分具有相关性。APOE基因型可能影响中老年人胆固醇水平与认知功能。
Objective To investigate the correlation between apolipoprotein(APO)E,APOA1 gene and fasting blood glucose(FBG),triglyceride(TG),cholesterol and Mini-Mental State Examination(MMSE)scores in middle-aged and elderly subjects.Methods From Jan.to Jun.2019,a total of 165 volunteers,receiving physical examination in Xiangtan Central Hospital were enrolled.MMSE scores,serum levels of FBG,TG and cholesterol were detected.Genomic DNA were extracted,and polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP)were used to analyze the polymorphism of APOE and APOA1 gene and their correlation with blood biochemical indicators and MMSE scores were analyzed.Results The cholesterol level ofε4 carriers in APOE gene was higher than that of non-ε4 carriers(P=0.017),but the MMSE scores were lower than non-ε4 carriers(P=0.005).In APOA1+83 bp gene,cholesterol level of T carriers was lower than C carriers(P=0.032).FBG and TG levels were not correlated with variant genotypes(P>0.05).Conclusion There could be correlation among APOE,APOA1+83 bp gene polymorphisms and serum cholesterol.Genotype of APOE could be correlated with MMSE scores.APOE genotype might have an effect on cholesterol and cognition in middle-aged and elderly subjects.
