摘要 目的探讨TLR4的基因Asp299Gly和Thr399Ile的单核苷酸多态性(SNPs)与梅毒的关系,为阐明梅毒易感性提供依据。方法收集本院皮肤科及性病门诊普查确诊的120例梅毒患者为实验组,招募60例健康人为对照组,采集外周血,提取全血基因组,PCR扩增Asp299Gly、Thr399Ile目的DNA序列,比对分析位点突变频率。结果梅毒患者TLR4基因Asp299Gly片段中共有19个位点存在多态性。梅毒患者TLR4基因Thr399Ile片段中共有22个位点存在多态性。Asp299Gly片段中2498位点突变率较高,差异具有统计学意义(P<0.05)。结论梅毒患者TLR4的基因Asp299Gly片段和Thr399Ile片段突变率增加。梅毒患者TLR4基因Asp299Gly片段中2498位点A缺失差异差异具有统计学意义,表明TLR4基因序列2498位点A缺失与梅毒遗传易感性可能相关。 Objective To investigate the association between the single nucleotide polymorphism SNPs about Asp299 Gly and Thr399 Ile of TLR4 gene and the syphilis,provide the basis for susceptibility to clarify syphilis.Methods One hundred and twenty patients with syphilis confirmed by general survey in dermatology and STD clinics of our hospital were selected as experimental group and 60 healthy persons were recruited as control group.Gather the peripheral blood,extract genomic DNA from human blood,and Asp299 Gly and Thr399 Ile DNA sequence are amplificated by PCR at the two groups respectively.At last,compare and analysis the frequence between the two groups.Results In the Asp299 Gly fragments of TLR4 gene of syphilitics,it has 19 polymorphism loci.In the Thr399 Ile fragments of TLR4 gene of sypholitics,it has 22 polymorphism loci.In study,the mutation rate of 2498 site on Asp299 Gly fragments is deamatic high,with statistical significance(P<0.05).Conclusion The mutation rate in the fragments of Asp299 Gly and Thr399 Ile of TLR4 gene could increase for syphilitics.It has statistical significance that the A is found deleted on 2498 site of TLR4 gene for syphilitics.It indicated that the A deletion on2498 site of TLR4 gene is associated with the genetic susceptibility to syphilis.
