摘要
目的探讨皖南地区汉族人群HLA-B基因单核苷酸多态性(Single Nucleotide Polymorphisms,SNP)rs34933313与肾综合征出血热(Hemorrhagic fever with renal syndrome,HFRS)的相关性。方法采集HFRS患者14例(病例组)和健康组50例(对照组)新鲜血样,提取基因组DNA,通过等位基因特异引物-聚合酶链反应(ASP-PCR)对HLA-B基因位点分型,应用SPSS22.0统计分析其等位基因及基因型两组间的差异。结果 HLA-B基因rs34933313位点受试人群分布符合哈迪-温伯格(Hardy-Weinberg,H-W)遗传平衡,两组等位基因G、C(χ~2=4.38,OR=2.45,P=0.04)和基因型GG、GC、CC(χ~2=6.47,P=0.04)频率分布差异均有统计学意义;病例组中C等位基因的分布频率高于G等位基因,且携带C等位基因罹患HFRS的概率是G等位基因的2.45倍;亚组分析发现HLA-B基因rs34933313位点在显性遗传模型下(χ~2=6.45,P=0.01,OR=10.21,95%CI=1.24~84.18)与HFRS发病风险具有相关。结论皖南地区汉族人群HLA-B基因rs34933313位点与HFRS发病风险存在相关性,且携带C等位基因者会提高HFRS的发生风险。
To investigate the relationship between single nucleotide polymorphisms(SNP) rs34933313 and Hemorrhagic fever with renal syndrome(HFRS) in the Han population of Southern Anhui Province. In this experiment, fresh blood samples from 14 HFRS patients(Case group) and 50 healthy patients(Control group) were collected and genomic DNA were extracted. HLA-B loci were classified by allele-specific primers polymerase chain reaction(ASP-PCR). The differences in alleles and genotypes between the two groups were in accordance with controlled by SPSS 22.0. The results showed that the distribution of rs34933313 of HLA-B gene in the groups was flowing Hardy Weinberg(H-W) genetic balance, and the differences of allele G、C(χ~2=4.38,OR=2.45,P=0.04) and genotype GG、GC、CC(χ~2=6.47,P=0.04) frequency distribution between the two groups were statistically significant, The distribution frequency of C allele in the case group was higher than that of G allele, and the probability of HFRS with C allele was G allele. The results of subgroup analysis showed that the rs34933313 locus of HLA-B gene was associated with the risk of HFRS in the dominant genetic model(χ~2=6.45, P=0.01, OR=10.21, 95%CI=1.24~84.18). It is suggested that the rs34933313 locus of HLA-B gene is related to the risk of HFRS in the Han population of Southern Anhui Province, and the risk of HFRS will be increased in C allele.
