基于系统药理学分析黄连-吴茱萸药对治疗胃肠道疾病 被引量:14

Analysis of the Mechanism of Rhizoma Coptidis-Fructus Evodiae Couplet Medicines in the Treatment of Gastrointestinal Diseases Based on Systematic Pharmacology

摘要 目的:运用系统药理学方法,整合中医药大数据,探讨黄连-吴茱萸药对治疗胃肠道疾病的作用机制。方法:在中药系统药理学分析平台(platform for systematic pharmacology analysis of traditional Chinese medicine,TCMSP)分别检索“黄连”“吴茱萸”获得其化学成分,以口服生物利用度(bioavailability,OB)、类药性(drug-likeness,DL)、Caco-2渗透率为指标筛选其活性成分,并通过文献检索补充不满足上述条件的黄连-吴茱萸药对活性成分或质量标志物;采用TCMSP、PharmMapper、GAD、TTD等数据库获取成分和胃肠道疾病相关靶标;运用生物学信息注释数据库(biological information annotation database,DAVID)对靶标进行通路注释和功能富集分析。构建成分、靶标、疾病、通路相互作用网络并进行可视化分析。结果:通过数据库和文本挖掘共筛选得到黄连-吴茱萸药对中与胃肠道疾病相关成分38个,涉及PTGS1、PTGS2、NOS2A、SLC64A、F2、MMP3等42个作用靶标,通过调控炎症性肠病、癌症通路、细胞因子-细胞因子受体相互作用、肿瘤坏死因子信号通路等20条信号通路发挥治疗胃肠道疾病的作用机制。结论:黄连-吴茱萸药对治疗胃肠道疾病具有多通路、多靶点的特点,为该药对及其相关中药制剂的研究提供思路和参考。 Objective:To study the action mechanism of Huanglian(Rhizoma Coptidis)-Wuzhuyu(Fructus Evodiae)couplet medicines in treatment of gastrointestinal diseases based on the systemic pharmacology and the big data of Chinese medicine.Methods:In the platform for systematic pharmacology analysis of traditional Chinese medicine(TCMSP),the chemical components of Huanglian(Rhizoma Coptidis)and Wuzhuyu(Fructus Evodiae)were searched respectively.The active components were screened by oral bioavailability(OB),drug-likeness(DL)and Caco-2 permeability.Retrieval of active ingredients or quality markers of Huanglian(Rhizoma Coptidis)-Wuzhuyu(Fructus Evodiae)couplet medicines that do not meet the above conditions through literature search.TCMSP,PharmMapper,GAD,TTD and other databases were used to obtain components and gastrointestinal disease related targets;Biological Information Annotation Database(DAVID)was used to conduct pathway annotation and functional enrichment analysis of targets.The interaction network of components,targets,diseases and pathways was constructed and visualized.Results:Through database and text mining,38 components related to gastrointestinal diseases in Huanglian(Rhizoma Coptidis)-Wuzhuyu(Fructus Evodiae)couplet medicines were screened,involving 42 targets such as PTGS1,PTGS2,NOS2A,SLC64A,F2 and MMP3,etc.and 20 signal pathways such as inflammatory bowel disease,cancer pathway,cytokine-cytokine receptor interaction,TNF signal pathway were regulated to play a role in the treatment of gastrointestinal diseases.Conclusion:Huanglian(Rhizoma Coptidis)-Wuzhuyu(Fructus Evodiae)couplet medicines has the characteristics of multi-channel and multi-target in the treatment of gastrointestinal diseases,which provides ideas and references for the study of Huanglian(Rhizoma Coptidis)-Wuzhuyu(Fructus Evodiae)couplet medicines and its related Chinese medicine preparations.
