摘要
目的:建立高效液相色谱(HPLC)波长切换法同时测定舒尔经胶囊中氧化芍药苷、芍药内酯苷、芍药苷、α-香附酮、延胡索乙素、紫堇碱和四氢小檗碱的含量。方法:采用HPLC,Agilent TC-C18色谱柱(250 mm×4.6 mm,5μm);流动相:乙腈-0.1%冰醋酸溶液,梯度洗脱,体积流量为1.0 mL·min^-1;柱温为30℃;检测波长分别为230 nm(检测氧化芍药苷、芍药内酯苷和芍药苷)、242 nm(检测α-香附酮)和280 nm(检测延胡索乙素、紫堇碱和四氢小檗碱)。结果:氧化芍药苷、芍药内酯苷、芍药苷、α-香附酮、延胡索乙素、紫堇碱和四氢小檗碱分别在4.29~85.80、17.07~341.40、25.99~519.80、3.36~67.20、1.47~29.40、1.19~23.80、0.96~19.20μg·mL-1线性关系良好(r≥0.9991),平均加样回收率分别为98.79%、99.54%、100.04%、97.94%、98.20%、97.75%和97.15%,RSD分别为1.19%、0.87%、0.79%、1.27%、1.35%、0.96%和1.54%。结论:建立的含量测定方法稳定、灵敏度高、重复性好,可用于舒尔经胶囊的质量评价。
Objective:To establish a HPLC multiwavelength switching method for simultaneous determination of oxypaeoniflorin,albiflorin,paeoniflorin,α-cyperone,tetrahydropalmatine,corydaline and tetrahydroberberine in Shu'erjing Capsules.Methods:HPLC was used on an Agilent TC-C18 chromatographic column(250 mm×4.6 mm,5μm),with acetonitrile-0.1%ice acetic acid solution as a mobile phase in gradient elution,at a flow rate of 1.0 mL·min^-1.The column temperature was 30℃.The detection wavelength were set at 230 nm for oxypaeoniflorin,albiflorin and paeoniflorin,242 nm forα-cyperone,280 nm for tetrahydropalmatine,corydaline and tetrahydroberberine.Results:Oxypaeoniflorin,albiflorin,paeoniflorin,α-cyperone,tetrahydropalmatine,corydaline and tetrahydroberberine showed good linear relationships within the ranges of 4.29-85.80,17.07-341.40,25.99-519.80,3.36-67.20,1.47-29.40,1.19-23.80,0.96-19.20μg·mL-1(r≥0.9991),respectively,whose average recoveries were 98.79%,99.54%,100.04%,97.94%,98.20%,97.75%,97.15%with the RSDs of 1.19%,0.87%,0.79%,1.27%,1.35%,0.96%,1.54%,respectively.Conclusion:The method was steady with high precision and good sensitivity.It could be used for the quality control in Shu'erjing Capsules.
