复方芪芎颗粒对佐剂型关节炎大鼠血清、滑膜液中IL-12、IL-18的影响 被引量:2

Effects of Compound Qixiong Granules on IL-12 and IL-18 in Serum and Synovial Fluid of Adjuvant Arthritis Rat

摘要 目的探讨复方芪芎颗粒对佐剂型关节炎(adjuvant arthritis,AA)大鼠血清、滑膜液中白细胞介素12(interleukin-12,IL-12)、IL-18的影响。方法将40只雄性Wistar大鼠随机分成正常对照组、模型对照组、复方芪芎组和甲氨喋呤组,每组10只。模型对照组、复方芪芎组和甲氨喋呤组大鼠通过弗氏完全佐剂诱导,建立AA模型。造模2周后进行给药:正常对照组和模型对照组大鼠给予0.9%氯化钠溶液灌胃;复方芪芎组大鼠给予复方芪芎颗粒药液(1 ml含生药1 g)灌胃;甲氨蝶呤组大鼠按体质量10 mg/kg的剂量给予甲氨蝶呤含药液灌胃;所有灌胃剂量均为10 ml/(kg·d),1次/d,持续3周。于造模后2周、给药后3周观察大鼠患肢足跖肿胀情况,并测量记录足跖厚度;连续治疗3周后,检测大鼠血清、滑膜液中IL-12、IL-18的表达量。结果造模大鼠均表现出足跖肿胀,血清、滑膜液中IL-12、IL-18表达量均高于正常对照组(P均<0.01)。治疗3周后,与模型对照组大鼠比较,复方芪芎组和甲氨喋呤组足跖肿胀度均减轻(P均<0.05),血清、滑膜液中IL-12、IL-18表达均下降(P均<0.05),且甲氨喋呤组效果优于复方芪芎组(P均<0.05)。结论复方芪芎颗粒能显著改善AA大鼠的关节炎症,其机制可能与抑制血清和滑膜液中IL-12、IL-18的表达有关。 Objective To explore the effect of compound Qixiong granules on interleukin-12(IL-12) and IL-18 in serum and synovial fluid of adjuvant arthritis(AA) rats.Methods Forty male Wistar rats were randomly divided into normal control group, model control group, compound Qixiong group and methotrexate group, with 10 rats in each group. Rats in the model control group, compound Qixiong group and methotrexate group were induced by Freund’s complete adjuvant to establish AA models. Dosing 2 weeks after modeling: rats in the normal control group and the model control group were given oral gavage with 0.9% sodium chloride solution;rats in the compound Qixiong group were given gavage of compound Qixiong granules(1 ml containing crude medicine 1 g);Rats in the methotrexate group were given methotrexate at a dose of 10 mg/kg of body weight;all gavage doses were 10 ml/(kg·d), once a day, for 3 weeks.The metatarsal swelling was observed at 2 weeks after modeling and 3 weeks after gavage, and the metatarsal thickness was measured and recorded. The IL-12 and IL-18 expressions in serum and synovial fluid of rats were dectected after 3 weeks of continuous treatment.Results The metatarsal swelling, and the expressions of IL-12 and IL-18 in serum and synovial fluid of the model rats were higher than those in the normal control group(all P<0.05). After 3 weeks of treatment, compared with the model control group, the metatarsal swelling of compound Qixiong group and methotrexate group were reduced(all P<0.05),the expressions of IL-12 and IL-18 in serum and synovial fluid were decreased(all P<0.05);and the effect of methotrexate group was better than that of compound Qixiong group(all P<0.05).Conclusion Compound Qixiong granules can significantly improve the arthritis in AA rats, and its mechanism may be related to the inhibition of IL-12 and IL-18 expressions in serum and synovial fluid.
