摘要
采用RT-PCR方法对从内蒙古呼和浩特地区采集的29份表现花叶症状的海红果(Malus micromalus Makino)叶片样品进行了苹果坏死花叶病毒(ApNMV)的检测,ApNMV的检出率为100%,说明ApNMV在呼和浩特地区表现花叶症状的海红果上普遍发生。分别克隆并测序了29份海红果样品中ApNMV的全长外壳蛋白(CP)基因,结果表明,29个ApNMV分离物CP基因间核苷酸序列一致性为94.1%~99.8%,与NCBI数据库中其他19个ApNMV分离物全长CP基因核苷酸一致性为86.1%~97.1%。利用ApNMV CP基因的全长序列构建了系统发育树,48个ApNMV分离物共分成5个组,本研究中的29个ApNMV分离物与2个日本分离物LC108995和NC040470聚集在一起,形成组Ⅰ;其余ApNMV分离物分别形成了组Ⅱ~组Ⅴ。
Malus micromalus Makino plants exhibiting the main symptom of mosaic leaves were observed in Hohhot. Fresh leaf samples from^29 symptomatic trees were collected for RT-PCR test of Apple necrotic mosaic virus(ApNMV) infection. Coat protein(CP) genes of ApNMV were amplified from all the collected samples. The nucleotide sequence identity among the 29 CP genes ranged 94.1% to 99.8%, while the identity between the 29 and another 19 ApNMV isolates published in NCBI GenBank was from 86.1% to 97.1%. A phylogenetic tree was reconstructed for ApNMV based on the CP gene sequences of 48 isolates, showed that the 29 isolates clustered together with another 2 isolates from Japan(GenBank accession no. LC108995 and NC040470), forming the Group Ⅰ;the other 17 isolates formed clusters of Group Ⅱ to Ⅴ.
