摘要
目的观察心脑宁胶囊对心肌梗死模型大鼠主动脉内膜结构、主动脉组织蛋白酶S(CatS)、沉默信息调节因子2相关酶1(SIRT1)表达的影响,探讨其稳定动脉粥样硬化斑块机制。方法 60只雄性SD大鼠,随机选出15只为假手术组,其余45只高脂饲料喂养6周,再进行冠脉结扎手术,造模成功后将存活大鼠随机分为模型组,西药组,中药组,继续高脂饲料喂养至8周。假手术组大鼠予正常饮食,西药组和中药组分别予阿托伐他汀钙和心脑宁胶囊灌胃,假手术组和模型组均予等量蒸馏水。用药4周后,HE染色法观察大鼠主动脉内膜结构变化,检测血清甘油三酯(TG)、胆固醇(TC)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)含量,Western Blot法检测主动脉内CatS、SIRT1含量变化。结果与假手术组相比,模型组大鼠动脉结构紊乱,内膜有斑块形成;血清TG、TC、LDL和主动脉Cat S含量升高,HDL和SIRT1含量减少(P<0.05)。与模型组比较,中药组及西药组大鼠动脉内膜较为平滑,血清TG、TC、LDL和主动脉CaS表达降低,HDL和APN含量升高(P<0.05);中药组与西药组间各指标差异无统计学意义(P>0.05)。结论心脑宁胶囊具有良好的稳定动脉粥样硬化斑块作用,推测与其改善动脉结构,调控动脉内CatS、SIRT1蛋白表达有关。
Objective To observe the impacts of Xinnaoning Capsules on the aortic intimal structure, expressions of CatS and SIRT1 in rats with myocardial infarction, and explore the mechanisms of Xinnaoning Capsules stabilizing atherosclerotic plaques. Methods Fifteen male SD rats in sham operation group were randomly selected from 60 rats and the rest 45 rats were fed with high fat diet for 6 weeks and received coronary artery ligation operation. The successful modeling rats were randomly divided into model group, western medicine group and TCM group, and were fed with high fat diet for 8 weeks. The sham operation group was fed with normal diet. The western medicine group and TCM group received atorvastatin and Xinnaoning Capsules, respectively. The sham operation group and model group received the same amount of distilled water. After 4 weeks treatment, the aortic intimal structure was observed by HE. The serum levels of TG, TC, LDL and HDL were measured. CatS and SIRT1 expressions of aorta were determined by Western Blot, respectively. Results Compared with the sham operation group, aortic intimal structure derangement and plaque formation in intima, serum levels of TG, TC, LDL and CatS increased,and HDL and APN decreased(P<0.05). Compared with model group, the endarterium was more smooth, serum levels of TG, TC, LDL and CatS decreased,and HDL and SIRT1 increased(P<0.05). And there were no significant differences between western medicine group and TCM group(P>0.05). Conclusion Xinnaoning Capsules showed good effects for stabilizing atherosclerotic plaques, partly by improving aortic intimal structure and modulating the expressions of CatS and SIRT1.
