摘要
目的:建立高效液相色谱(HPLC)法测定甘草银花炙草乌单酯型生物碱的含量测定及双酯型生物碱的限量检查方法,为甘草银花炙草乌的质量控制提供科学依据。方法:色谱柱Hydrosphere C18(4.6 mm×250 mm,5μm),以乙睛-四氢呋喃(25∶15)为流动相A,以0.1 mol·L-1醋酸铵溶液(每1000 mL加冰醋酸0.5 mL)为流动相B,梯度洗脱,流速1.0 mL·min-1,检测波长235 nm,柱温为25℃。结果:相邻成分色谱峰分离度较好,阴性无干扰。各化合物的浓度与峰面积的线性关系良好(r≥0.995),9批样品单酯型生物碱的含量范围为0.032%~0.051%,双酯型生物碱的含量范围为0.0001%~0.0031%。结论:该方法结果准确、稳定、可靠,可以用于甘草银花炙草乌的质量依据,可有效控制产品质量。
Objecetive:To establish methods for determination content of monoester alkaloids,and for the limit of diester alkaloids,to provide scientific basis for quality control of Licorice honeysuckle broiled aconite.Methods:The method was performed on a Hydrosphere C18(4.6 mm×250 mm,5μm)column,and acetonitrile-tetrahydrofuran(25∶15)as mobile phase A,0.1 mol·L-1 ammonium acetate solution as mobile phase B.The flow rate was 1.0 mL·min-1 and the detection wavelength was 235 nm.The column temprerature was 25℃.Results:The separation degree was high between each components,and the negative control has no interference.The linear relationship between concentration and peak area of each compound is good(r≥0.995).The content range of monoester alkaloids is 0.032%-0.051%,and the content range of diester alkaloids is 0.0001%-0.0031%.Conclusion:The HPLC method is fast,accurate and reproducible,which provides scientific basis for the quality control of Licorice honeysuckle broiled aconite.
