粒用高粱亲本系萌发期耐碱性鉴定与综合评价 被引量:2

Identification and evaluation of alkali tolerance of grain sorghum parental lines during germination

摘要 [目的]确定高粱萌发期耐碱鉴定适宜的NaHCO3∶Na2CO3=9∶1胁迫浓度、指标和评价方法,为大规模鉴定耐碱种质提供评价方法及进一步选育耐碱高粱新品种和发掘耐碱基因奠定基础。[方法]首先以10份粒用高粱亲本系为材料,在不同浓度的NaHCO3∶Na2CO3=9∶1(0、25、50、75、100、150、200 mmol·L^-1)胁迫下进行发芽试验,通过对发芽势、发芽率、发芽指数、芽长和根长进行统计和方差分析,筛选出鉴定粒用高粱萌发期耐碱性最适宜的NaHCO3∶Na2CO3=9∶1胁迫浓度。后续以50 mmol·L^-1 NaHCO3∶Na2CO3=9∶1溶液为胁迫浓度,对146份粒用高粱亲本系萌发期进行耐碱性鉴定,测定发芽率、中胚轴长、胚芽长、胚根干重等11个萌发性状,并进行主成分分析。[结果]通过平均隶属函数值S值制定耐碱分级标准,评价146份粒用高粱亲本系的耐碱性。根据S值分级标准把146份亲本系按耐碱性强弱分为5级:吉4327R为强耐碱类型,MH等8份为耐碱类型,LT等62份为中等耐碱类型,2483等56份为碱敏感类型,龙188B等19份为碱极敏感类型。主成分分析结果表明,相对中胚轴长、相对芽鲜重和相对根鲜重分别在3个主成分中的负荷量最大,且以其平均隶属函数值划分的耐碱评价结果与S值基本一致,相对中胚轴长、相对芽鲜重和相对根鲜重这3个指标可以作为粒用高粱萌发期耐碱性鉴定的适宜指标。[结论]高粱亲本系萌发期耐碱(NaHCO3∶Na2CO3=9∶1)鉴定的适宜胁迫浓度为50 mmol·L^-1,制定了高粱萌发期耐碱级别划分标准,筛选出萌发期强耐碱种质吉4327R。 [Objective]The aims of this study were to determine appropriate stress concentration of NaHCO3∶Na2CO3 solution at 9∶1 ratio(NNS91),the major stress-response indexes,and the evaluation method for alkali tolerance identification during sorghum germination,to provide an evaluation method for large-scale identification of alkali germplasm,and to lay the foundation for exploring alkali genes and breeding new sorghum varieties.[Methods]The seed germination test was set up with 10 sorghum parent lines which were individually treated with NNS91 at the concentration of 0,25,50,75,100,150,and 200 mmol•L^-1.Statistical and variance analysis results on germination force,germination rate,germination index,embryo length,and root length of seedling demonstrated that 50 mmol•L^-1 was the most suitable stress concentration of NNS91 to produce significant difference on different germinating parameters for grain sorghum parental lines.Therefore,50 mmol•L^-1 of NNS91 was applied to 146 sorghum parental lines to test their alkali resistant level,and the 11 germination traits including germination rate,mesocotyl length,embryo length,and root dry weight of seedling were measured and principal component analysis was performed.[Results]Alkali-resistant grading standards were established through average membership function value S to evaluate the alkali resistant levels of all tested material.According to the S value grading standard,146 copies of the parental line were divided into five grades based on the alkali resistance:Ji 4327R was a strong alkali resistance type;MH and other seven material were belong to alkali resistance type;LT and other 61 were medium alkali resistance type;56 material including 2483 were alkali sensitive type,and the rest of 19 including Long 188B were alkali super sensitive type.The principal component analysis showed that the relative mesocotyl length,embryo fresh weight,and root fresh weight were the most important contributors to three principal components in that order.The evaluation results of alkali tolerance divided by their average membership function values were consistent with the S value.Therefore,these three indexes could be used as suitable indicators for the identification of alkali tolerance during germination of sorghum grains.[Conclusion]In this study,50 mmol•L^-1 was determined as the optimal NNS91 concentration for alkali tolerance screening.In addition,the standard of alkali tolerance classification during sorghum germination was established,and the highly alkali tolerance parental line Ji 4327R was screened out.
