摘要
目的建立复方五凤草液HPLC指纹图谱与复方五凤草液总黄酮含量的测定方法,为全面评价复方五凤草液质量提供方法。方法采用HPLC法建立指纹图谱,对主要指纹峰进行了归属。同时以芦丁为对照品,用三氯化铝法测定复方五凤草液中总黄酮含量,对方法测定条件进行系统优化,并进行方法学验证。结果 HPLC指纹图谱方法精密度、重复性和稳定性良好。总黄酮含量测定最佳条件为检测波长415nm,0.1mol·L^-1显色剂用量为2mL,pH 5.2的乙酸-乙酸钠缓冲液用量为1mL,显色时间为30min。芦丁标准品在0.006~0.030mg·mL^-1质量浓度范围内,吸光度与质量浓度呈良好的线性关系,相关系数r=0.9998,平均加样回收率为105.75%,RSD为3.15%。结论建立的HPLC指纹图谱与总黄酮测定方法简单易操作、精确、专属性强、重复性好,为该复方的质量控制提供方法依据。
OBJECTIVE Establish the HPLC fingerprint and a method to determine the content of flavonoids in Compound WuFengCao Liquid,providing a comprehensive method for evaluating the quality of compound WuFengCao liquid.METHODS The fingerprint was established by HPLC method and the main compounds were identified.At the same time,choosing rutin as a standard,determined the content of total flavonoids by aluminum trichloride method.Optimized the method determination conditions and finished the method validation.RESULTS The HPLC fingerprint method is suitable in precision,repeatability and stability.The optimum conditions for the determination of total flavonoids were 415 nm,the amount of aluminum trichloride developer was 2 mL,the pH of 5.2 acetic acid-sodium acetate buffer was 1 mL,and the color development time was 30 minutes.The rutin standard showed a good linear relationship with the absorbance in the range of 0.006-0.03 mg·mL^-1,r=0.9998.The average recovery rate of the sample was 105.75%,and the RSD was 3.15%(n=6).CONCLUSION The established HPLC methods and determination of total flavonoids methods are simple,easy to operate,precise,specific,and reproducible,providing a theoretical basis for the quality control of the compound.
