基于网络药理学分析越鞠丸治疗抑郁症的分子机制 被引量:9

摘要 目的通过分析越鞠丸的有效化学成分,研究其治疗抑郁症的分子机制。方法将越鞠丸中的5味中药通过中药系统药理学技术平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)数据库检索化学活性物质,筛选出药物作用于人体的靶点蛋白。应用基因组注释数据库平台(Genecards)预测抑郁症的作用靶点。将所得活性成分与靶点应用uniprot数据库查询出相对应的基因名称,形成活性成分-靶点网络图。应用String数据库平台形成蛋白互作网络,然后分别通过Bioconductor平台、R语言GO富集分析、KEGG富集分析,形成靶点-通路网络图,分析越鞠丸治疗抑郁症的分子机制。结果越鞠丸治疗抑郁症的关键化学成分主要有26个,共同靶点90个,通过PPI蛋白互作关系,得到15个核心基因,分别为MAPK1、AKT1、HSP90AA1、IL6、FOS、MAPK14、MAPK8、ESR1、NR3C1、STAT1、VEGFA、EGF、IL1B、AR、IL2;GO富集分析结果包括调节神经递质水平、营养水平应答、膜筏、儿茶酚胺结合等;KEGG富集通路包括AGE-RAGE信号通路、IL-17信号通路、TNF信号通路等。结论越鞠丸可通过调节MAPK10、MAPK8、AKT1、MAPK1、MAPK14、FOS、IL6、CASP8、CASP3等靶点基因,调控AGE-RAGE信号通路、IL-17信号通路、TNF信号通路等来实现调节神经递质水平、神经保护,从而起到抗抑郁作用。 Objective To analyze the effective chemical composition of Yueju pills and study its molecular mechanism for depression.Methods The five Chinese medicines in Yueju pill were searched for chemically active substances through the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)database to screen out the target proteins of the drugs acting on the human body.Using the genome annotation database platform(Genecards)to predict the target of depression.The obtained active ingredients and targets were queried by uniprot database to find out the corresponding gene names to form an active ingredient-target network diagram.The string database platform was used to form a protein interaction network,and then through the bioconductor platform,R language GO enrichment analysis,and KEGG enrichment analysis to form a target-pathway network diagram to analyze the molecular mechanism of Yueju pills in the treatment of depression.Results There were 26 key chemical compo-nents for Yueju pill in the treatment of depression,and 90 common targets.Through PPI protein interaction,15 core genes were obtained,including MAPK1,AKT1,HSP90AA1,IL6,FOS,MAPK14,MAPK8,ESR1,NR3C1,STAT1,VEGFA,EGF,IL1B,AR,IL2;GO enrichment analysis results included regulation of neurotransmitter levels,nutritional level re-sponse,membrane raft,catecholamine binding,etc.KEGG enrichment pathway included AGE-RAGE signaling path-way,IL-17 signaling pathway,TNF signaling pathway,etc.Conclusion Yueju pill can regulate the neurotransmitter level and has neuroprotection and antidepression effect by regulating target genes,including MAPK10,MAPK8,AKT1,MAPK1,MAPK14,FOS,IL6,and CASP3,and regulating the signaling pathway of AGE-RAGE,IL-17 and TNF signaling pathway.
