摘要
目的探究慢性乙型肝炎病毒(HBV)感染患者细胞免疫与miRNA-155表达的相关性。方法选取2018年2月到2019年2月我院收治的慢性HBV患者40例设为观察组,选取同期入院检查的健康个体40例设为对照组。采集所有患者外周血单个核细胞,并测定其微小核糖核酸-155(miRNA-155)表达量(xMAP液态芯片技术),同时测定所有人员外周血T细胞亚群,包括CD3^+百分率,CD4^+百分率,CD8^+百分率,CD4^+/CD8^+比值,对miRNA-155与T细胞亚群指标进行相关性分析。结果观察组CD3^+、CD4^+以及CD4^+/CD8^+明显低于对照组,且miRNA-155相对表达量明显低于对照组,差异具有统计学意义(P<0.05);CD8^+百分率两组相比无明显差异,不具统计学意义(P>0.05)。以miRNA-155相对表达量作为因变量,以T细胞亚群各指标含量作自变量,进行多因素线性回归分析,结果显示:CD3^+百分率与miRNA-155呈正相关(B=0.050,P=0.000,95%CI=0.037-0.064),其余指标与miRNA-155并未表现出相关性(P>0.05)。以观察组与对照组miRNA-155相对表达量作为区分标准,建立状态变量(观察组患者miRNA-155=0,对照组miRNA-155=1),同样将所有人员T细胞亚群各指标含量作自变量,进行多因素Logistics回归分析,结果显示:CD3^+百分率与miRNA-155表现出相关性(B=0.601,P=0.008,95%CI=1.171-2.838),其余指标与miRNA-155并未表现出相关性(P>0.05)。结论慢性HBV感染患者的miRNA-155表达量相较于正常人群明显更低,这与其细胞免疫能力的抑制表现出了相关性,但该研究结果仅仅发现T细胞亚群中CD3^+百分率与miRNA-155表达量呈正相关,但本研究例数有限,无法确定其准确性,仅供参考。
Objective To explore the correlation between cellular immunity and mirna-155expression in patients with chronic hepatitis b virus(HBV)infection.Methods 40patients with chronic HBV admitted to our hospital from February 2018to February 2019were selected as the observation group,and 40healthy individuals admitted to our hospital during the same period were selected as the control group.Peripheral blood mononuclear cells of all patients were collected and their expression level of microrbonucleic acid-155(mirna-155)was measured(xMAP liquid chip technology).Meanwhile,t-cell subsets in peripheral blood of all patients were measured,including CD3^+ percentage,CD4^+percentage,CD8^+ percentage,and CD4^+/CD8^+ ratio.Correlation analysis was conducted between mirna-155and t-cell subsets.Results CD3^+,CD4^+ and CD4^+/CD8^+ in the observation group were significantly lower than those in the control group,and the relative expression level of mirna-155was significantly lower than that in the control group,with statistically significant differences(P<0.05).There was no significant difference in CD8^+ percentage between the two groups(P>0.05).The relative expression of mirna-155was taken as the dependent variable,and the content of each indicator in the T cell subpopulation was taken as the independent variable,and the multivariate linear regression analysis was conducted.The results showed that the percentage of CD3^+was positively correlated with mirna-155(B=0.050,P=0.000,95%ci=0.037-0.064),while the other indicators showed no correlation with mirna-155(P>0.05).The relative expression level of mirna-155in the observation group and the control group was used as the distinguishing criterion,and state variables were established(mirna-155=0in the observation group,and mirna-155=1in the control group).The contents of various indicators in all human T cell subsets were also used as independent variables,and multi-factor logistic regression analysis was conducted,and the results showed:The percentage of CD3^+ was correlated with mirna-155(B=0.601,P=0.008,95%ci=1.171-2.838),while the other indicators were not correlated with mirna-155(P>0.05).Conclusion The expression level of mirna-155in patients with chronic HBV infection is significantly lower than that in the normal population,which is correlated with the suppression of cellular immunity.However,this study only found that the percentage of CD3^+ in T cell subsets was positively correlated with the expression level of mirna-155.However,the number of cases in this study was limited,so the accuracy of this study could not be determined,which is only for reference.
