摘要
【目的】研究姜黄素对四氯化碳诱导的鲤(Cyprinus carpio)肝脏损伤的修复作用,为筛选治疗鱼类肝脏综合征的绿色药物提供理论依据。【方法】在基础饲料中分别添加0、60、120、240 mg/kg姜黄素,制备4种等氮、等能饲料。试验设5组,阴性对照组注射橄榄油,饲喂基础饲料,阳性对照组和各姜黄素组注射体积分数15%的CCl4溶液,分别以添加0、60、120、240 mg/kg姜黄素饲料饲喂14 d,测定各组谷丙转氨酶(GPT)、谷草转氨酶(GOT)活力,丙二醛(MDA)含量,血浆总蛋白(TP)含量,总能氧化能力(T-AOC),肝脏超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)活力,以及肝细胞核相关转录因子2(Nrf2)含量等指标,并对各组肝脏细胞进行组织学观察,研究姜黄素对鲤肝损伤的修复作用。【结果】在四氯化碳诱导初期,阳性对照组,60、120、240 mg/kg姜黄素组的GPT、GOT活力和MDA含量显著高于阴性对照组(P <0.05),随着投喂时间的增加,各姜黄素组GPT、GOT活力和MDA含量平均值均有不同程度的降低,其中,120 mg/kg组的GPT、GOT活力和MDA含量以及240mg/kg组的GPT活力差异显著(P<0.05);TP含量,T-AOC、SOD、GSH-Px活力,GSH含量的变化趋势与GPT和GOT等相反,且随着投喂时间的增加,120mg/kg组的T-AOC、SOD、GSH-Px活力和TP、GSH含量以及240 mg/kg组的SOD、GSH-Px活力和GSH含量呈先降后升趋势(P <0.05),其余各组差异均不显著(P> 0.05);肝脏组织切片显示,饲料中添加姜黄素可一定程度上修复鲤鱼幼鱼的肝脏损伤,且以120 mg/kg组效果最佳;饲料中添加姜黄素可提高鲤鱼肝脏Nrf2 mRNA表达量,促使Nrf2由肝细胞质转至细胞核中。【结论】姜黄素可通过介导Nrf2提高鲤鱼相关抗氧化酶的活力,对鱼体的肝脏损伤进行修复。当姜黄素的添加水平为120 mg/kg时,机体的抗氧化功能最强,对肝脏损伤的修复作用最强。
【Objective】To evaluate the recovery of curcumin in liver injury of Cyprinus carpio induced by carbon tetrachloride(CCl4),for future screening of green treatment drugs for the tr fish hepatobiliary syndrome.【Methods】A basal diet supplemented with 0,60,120 and 240 mg/kg curcumin as four experimental diets.The experiment included five groups:positive control group(injected with 15%carbon tetrachloride solution,fed basic feed),negative control group(injected with olive oil,fed basic feed),60,120 and 240 mg/kg curcumin group(injected with 15%carbon tetrachloride solution,fed the diet supplemented with 60,120 and 240 mg/kg curcumin respectively).The activity of GPT,GOT,MDA content,Total Protein(TP)in blood,T-AOC,SOD,GSH,GSH-Px,and expression of nuclear factor erythroid 2-related factor 2(Nrf2)were detected,and histological exaination for liver was conducted.【Results】By comparing blood physiological indexes(GPT,GOT,TP and T-AOC)and other antioxidant indexes(SOD,MDA,GPx,GSH and Nrf2,et al)among groups at different times,the therapy effect of curcumin to liver injury induced by carbon tetrachloride in Cyprinus carpio were evaluated.It was found that:during the experiment period,blood physiological indexes and other antioxidant indexes are significantly(P<0.05)influenced by the dose and feeding time of curcumin.Initially,there were a significant(P<0.05)increase on glutamic oxalacetic transaminase(GOT)and glutamic pyruvic transaminase(GPT)activities of the plasma and liver malondialdehyde(MDA)content in the positive control group and the three curcumin groups,and the mean values of these indexes decreased with increasing feeding time.The activity of GPT,GOT and MDA content of 120 mg/kg group and GPT activity of 240 mg/kg group had a significantly difference(P<0.05)with increasing feeding time.In addition,the trend of total protein(TP)and total antioxidant capacity(T-AOC)of plasma and liver superoxide dismutase(SOD),glutathione(GSH)and glutathione peroxidase(GSH-Px)activities were the opposite of that of GPT.T-AOC,SOD and GSH-Px activities and TP and GSH contents of 120 mg/kg group.The activity of SOD and GSH-Px and GSH content of 240 mg/kg group increased firstly and then decreased(P<0.05)with increasing feeding time.Curcumin(120 mg/kg curcumin per diet)can repair the damage of liver caused by carbon tetrachloride and restored liver tissue to normal.Results of Nrf2 mRNA expression in liver and Nrf2 protein level in nucleus also indicated that the dietary supplementation with optimal curcumin could increase the Nrf2 mRNA expression in liver and enhance antioxidant status of carp by promoting the release of Nrf2,which increase the activity of antioxidant enzymes and finally protect common carp from liver injury by CCl4.【Conclusion】Curcumin(especially 120 mg/kg)can improve activities of related antioxidant enzymes through the regulation of Nrf2 activity,so as to repair liver damage in fish.
