摘要
为探究不同消毒方法对茶苗发芽率及消毒效果的影响,以云南小叶种茶树"十里香"种子为试验材料,采用种子萌发的无菌试管苗叶片及茎段为外植体,研究不同培养基类型、激素种类及浓度对愈伤组织诱导、增殖的影响。结果表明:4%次氯酸钠10 min+75%酒精30 s的消毒效果最好,污染率最低;叶片愈伤组织诱导培养基和叶片愈伤组织继代培养基的适宜配方均为B5培养基+0.5 mg/L 2,4-二氯苯氧乙酸(2,4-D)+0.1 mg/L激动素(KT);茎段愈伤组织诱导培养基和茎段愈伤组织继代培养基适宜配方均为MS培养基+1.5 mg/L 6-苄氨基嘌呤(6-BA)+0.2 mg/L萘乙酸(NAA)。研究结果为云南小叶种茶树组织培养及遗传转化体系的建立提供了科学依据。
To explore the effects of different disinfection methods on the germination rate and disinfection effect of tea seedlings, the seed of "Shilixiang" of Yunnan lobular tea plant was used as experimental material. The leaves and stem segments of seed germination were used as explants to study the induction and proliferation of callus by different media types, hormone types and concentrations. Results show that 4% sodium hypochlorite 10 min+75% alcohol 30 s has the best disinfection effect and the lowest pollution rate. The most appropriate formula for leaf callus induction medium and leaf callus subculture medium is B5 medium+0.5 mg/L 2, 4-Dichlorophenoxyacetic acid(2, 4-D)+0.1 mg/L kinetin(KT). The suitable medium for callus induction medium and stem segment callus induction medium are MS medium+1.5 mg/L 6-Benzylaminopurine(6-BA)+0.2 mg/L Naphthaleneacetic acid(NAA). The research results provide a scientific basis for the tissue culture and genetic transformation system of Yunnan lobular tea plantation.
