摘要
目的探讨长链非编码RNA(LncRNA)-生长停滞特异性转录物5(GAS5)与微小RNA(miR)-103在浸润性乳腺癌中的表达关系及临床意义。方法收集72例浸润性乳腺癌经手术切除的癌组织及癌旁正常组织标本,实时荧光定量聚合酶链式反应法(qRT-PCR)测定LncRNA-GAS5、miRNA-103相对表达量,并收集患者病理资料,分析浸润性乳腺癌LncRNA-GAS5、miRNA-103表达与病理参数的关系及两者表达的相互关系。结果浸润性乳腺癌癌组织LncRNAGAS5相对表达量低于癌旁组织,miR-103相对表达量高于癌旁组织(P<0.05)。随临床分期的上升,LncRNA-GAS5表达降低,miR-103表达上升;伴淋巴结转移浸润性乳腺癌患者癌组织LncRNA-GAS5表达低于未伴淋巴结转移者,miR-103高于未伴淋巴结转移患者(P<0.05)。浸润性乳腺患者癌组织LncRNA-GAS5与miR-103表达呈负相关(P<0.05)。结论浸润性乳腺癌患者癌组织LncRNA-GAS5呈低表达,miR103呈高表达,两者表达呈负相关,且均与乳腺癌临床分期、淋巴结转移有关。
Objective To explore the expressions and clinical significance of long-chain non-coding RNA(LncRNA)-growth arrest-specific transcript 5(GAS5)and microRNA(miR)-103 in invasive breast cancer.Methods Cancer tissues and adjacent normal tissues samples surgically resected from 72 cases of invasive breast cancer were collected.The relative expression levels of LncRNA-GAS5 and miRNA-103 were determined by real-time fluorescent quantitative polymerase chain reaction(qRTPCR).Pathological data were collected to analyze the relationship between expressions of LncRNA-GAS5 and miRNA-103 and pathological parameters in invasive breast cancer and the interrelationship between the 2 expressions.Results The relative expression of LncRNA-GAS5 in invasive breast cancer tissues was lower than that in adjacent tissues while the relative expression of miR-103 was higher than that in adjacent tissues(P<0.05).With the increase of clinical stages,the expression of LncRNAGAS5 was decreased while the expression of miR-103 was increased.The expression of LncRNA-GAS5 in cancer tissues in invasive breast cancer patients with lymph node metastasis was lower than that in patients without lymph node metastasis while the expression of miR-103 was higher than that in patients without lymph node metastasis(P<0.05).LncRNA-GAS5 in cancer tissues in patients with invasive breast cancer was negatively correlated with miR-103 expression(P<0.05).Conclusion LncRNAGAS5 is lowly expressed and miR103 is highly expressed in patients with invasive breast cancer,and the 2 expressions are negatively correlated and are related to clinical stages and lymph node metastasis of breast cancer.
