摘要
实验旨在探究蛋鸭蛋白激酶Cα(PRKCA)基因多态性与蛋壳品质的关联性,以期找到与蛋壳品质相关的分子标记。利用PCR与直接测序相结合的方法对蛋鸭PRKCA基因外显子区域的SNP突变位点进行检测,并与蛋壳品质性状进行关联性分析。结果显示:在樱桃谷鸭第7外显子区域检测到2个SNP位点g.9583228G>A和g.9583222C>T,均为同义突变且均产生3种基因型。2个SNP位点之间强连锁不平衡,发现3种单倍型和6种双倍型,单倍型H1和双倍型H1H1频率最高;关联性分析结果表明:双倍型H1H3和H2H2个体蛋重显著高于H3H3个体;H2H3、H3H3双倍型个体蛋壳厚度显著高于H2H2个体;H3H3蛋壳强度显著高于H2H2个体。综上,双倍型H3H3是改善樱桃谷鸭蛋壳品质最有利的双倍型,揭示在PRKCA基因外显子7发现的2个SNP位点相互联合能显著影响蛋壳品质,可能作为鸭蛋壳品质选择的遗传标记。
PRKCA gene is a basic regulator of calcium transport, which plays an important role in regulating ion channels, cell secretion and exocytosis, cell growth and proliferation. The purpose of this study was to explore the association between protein kinase C-alpha(PRKCA) polymorphism and eggshell quality in laying ducks, in order to find molecular markers related to eggshell quality. Two SNP loci g.9583228 G>A and g.9583222 C>T were detected in exon 7 of PRKCA gene of laying ducks by PCR and direct sequencing. They were synonymous mutations and produced three genotypes. Strong linkage disequilibrium was found between two SNP loci, and three haplotypes and six doubles were found. The frequency of haplotypes H1 and doubles H1 H1 was the highest. Relevance analysis showed that egg weight of doubled H1 H3 and H2 H2 individuals was significantly higher than that of H3 H3(P<0.05);eggshell thickness of doubled individuals of H2 H3 and H3 H3 was significantly higher than that of H2 H2 individuals(P<0.05);The strength of eggshell was significantly higher than that of H2 H2(P<0.05). In conclusion, it can be seen that double H3 H3 is the most favorable double type for improving the eggshell quality of Cherry Valley ducks. It is revealed that the combination of two SNP loci found in exon 7 of PRKCA gene can significantly improve the eggshell quality, which may serve as a genetic marker for the selection of duck eggshell quality.
