摘要
[目的]为了探明EuREF1与胶分子量和含胶率的关系,深入探究杜仲胶合成机制。[方法]在杜仲新枝和叶片生长较快的4月中旬到6月中旬,利用RT-PCR技术分析杜仲EuREF1基因在雌雄株叶片和茎皮中的表达水平,以索氏提取法和GPC/SEC分别检测叶片和树皮中橡胶的含量和分子量。[结果]在4月至6月期间,雌雄株叶片、茎皮EuREF1表达水平分别与这些器官中胶含量及胶分子量呈显著或极显著正相关(R=0.8982~0.9880)。在雌株叶片、茎皮、果皮和雄株叶片、茎皮中,分子量在1.0×10^6~5.0×10^6 Da所占比例最多,即长度在1.5×10^4~7.4×10^4个异戊二烯单体的橡胶最多。[结论]结果推测EuREF1基因产物与杜仲胶积累程度有紧密的关系,可能在橡胶链的延伸中发挥重要作用。
[Objective]To study the relationship between EuREF1 and molecular weight and rubber content,and to understand the synthesis mechanism of gutta-percha.[Method]In mid-April to mid-June when the new stems and leaves of Eucommia ulmoides grew rapidly,RT-PCR technique was used to analyze the expression level of EuREF1 gene in male and female leaves and stems,and Soxhlet extraction and GPC/SEC were used respectively to detect the rubber content and molecular weight of rubber in leaves and stems.[Result]From April to June,the expression levels of EuREF1 in female leaves,stems and male leaves,stems were significantly or extreme-significantly positively correlated with the rubber content and molecular weight in these plant organs(R=0.8982-0.9880).Among the leaves,stems,fruit of female and leaves,stems of male,the proportion of molecular weight of 1.0×10^6 Da to 5.0×10^6 Da was the highest,and the rubber with length of 1.5×10^4 to 7.4×10^4 isoprene residues was the most.[Conclusion]The results suggest that the product of EuREF1 gene is closely related to the accumulation of gutta-percha and may also plays an important role in the elongation of rubber chain.
